| de |

dc.description.abstracteng | Mycophenolic acid (MPA), a widely used immunosuppressive agent, exerts its pharmakological action through uncompetitive inhibition of inosine monophosphate dehydrogenase type II (IMPDH-II), a key enzyme in de novo synthesis of purine nucleotides. Mycophenolic acid is metabolised in vivo to a pharmakologically inactive phenolic glucuronide (MPAG) and an pharmakologically active acyl glucuronide (AcMPAG). Investigations on the inhibition of IMPDH-II by AcMPAG showed that AcMPAG is like MPA an uncompetitive inhibitor of IMPDH-II with a Ki that is around 6-10 fold higher than that of MPA and that binding to human serum albumin reduces this inhibition. | de |
