dc.description.abstractDie vorliegende Arbeit setzt sich mit der Frage auseinander, inwieweit Apoptose- und Proliferationsereignisse zur Morphogenese von Somiten beitragen. Untersucht wurden für die strukturelle Feinanalyse besonders geeignete Semidünnschnittserien von 12 bis 16 Tage alten Embryonen der primatennahen Tupaia belangeri. Das verwendete Scan- und Rekonstruktionssystem wurde in Vorarbeiten etabliert. Die in der Literatur geäußerte Vermutung, dass die Abgliederung von Somiten aus dem paraxialen Mesoderm regelhaft durch Apoptoseprozesse unterstützt würde, können wir nicht bestätigen. Die Bildung intersomitischer Spalten geht bei Tupaia belangeri von zwei einander gegenüber liegenden Spaltinitialisierungszonen aus, deren erstmaliger Nachweis in ventromedialen und dorsolateralen Abschnitten des paraxialen Mesoderms bestehende widersprüchliche Auffassungen zum Ablauf der Spaltbildung relativieren könnte. Schließlich haben wir Kandidaten für Grenzzellen identifiziert, deren Existenz und Spalt-bildende-Funktion auf der Grundlage von Wachtel-Huhn-Transplantationsexperimenten postuliert worden ist. Mit Hilfe dreidimensionaler Rekonstruktionen konnten wir die Sequenz der epithelial-mesenchymalen Transition von Somitenzellen ins Sklerotom genauer als bisher bekannt dokumentieren. Demnach setzt die Sklerotombildung medial ein, schreitet hier zunächst nach kranial und kaudal fort, und dehnt sich erst dann nach lateral aus. Apoptosehäufungen innerhalb des Sklerotoms treten, wie bei Haushuhn und Labormaus, gehäuft in dessen lateraler Subdomäne auf. Ein neuer Befund ist die Überlappung von Apoptose- und Proliferationsmaxima in der ventralen Subdomäne des Sklerotoms, die mit der Bildung der Perichordalscheide in Zusammenhang stehen könnten. In der zentralen, zwischen Neuralrohr und Myotom lokalisierten Subdomäne steht ein weiteres Proliferationsmaximum einem Apoptoseminimum gegenüber. Bei Tupaia belangeri stammen die Vorläuferzellen epaxialer Myotome aus der dorsomedialen Lippe, die des hypaxialen Myotoms aus der ventrolateralen Lippe des Dermomyotoms. Die De-epithelialisierung der Dermomyotome beginnt im Rumpfbereich zentral, in Segmenten dagegen, die kein hypaxiales Myotom ausbilden, in der ventrolateralen Lippe. Proliferationsmaxima unter Vorläuferzellen der Myotome fanden wir zunächst isoliert in der dorsomedialen und ventrolateralen Lippe, dann in allen vier Lippen des Dermomyotoms und schließlich diffus im gesamten Dermomyotom. Apoptosemaxima, die während der De-epithelialisierung in der dorsomedialen Lippe sowie in zentralen Abschnitten der Dermomyotome des Rumpfbereichs auftreten, nehmen offenbar eine Selektion unter Vorläuferzellen der Dermis vor. Auch der Bildung hypaxialer Myotome gehen Apoptosehäufungen in der ventrolateralen Lippe voraus. Dagegen werden Vorläuferzellen der Extremitätenmuskeln möglicherweise erst während ihrer Wanderung zu den Zielgebieten apoptotisch selektioniert. Innerhalb der Myotome nimmt die Zahl mitotischer und apoptotischer Zellen, wie bei anderen Wirbeltieren, mit dem Entwicklungsfortschritt zu. Auch die präferenzielle Ausbildung von Proliferationsmaxima in dorsomedialen und von Apoptosemaxima in ventrolateralen Myotomabschnitten ist offenbar eine Konstante bei Vertretern aller Wirbeltierklassen. Simultanrekonstruktionen des paraxialen und intermediären Mesoderms, des Seitenplattenmesoderms sowie der Chorda dorsalis von Tupaia belangeri erbrachten den Befund, dass bandförmig angeordnete Apoptoseereignisse offenbar eine Grenzziehung zwischen dem Seitenplattenmesoderm und den medial benachbarten Mesodermabschnitten vornehmen. Außerdem postulieren wir die Existenz pro-apoptotischer Faktoren im Seitenplattenmesoderm, die benachbarte Apoptosehäufungen (1) in der lateralen Subdomäne des Sklerotoms, (2) in der ventrolateralen Lippe von Dermomyotomen des Rumpfes, (3) unter migrierenden, aus der ventrolateralen Lippe des Dermomyotoms stammenden Vorläuferzellen der Extremitätenmus keln sowie (4) in der ventrolateralen Hälfte epaxialer Myotome verursachen könnten. Am kaudalen Embryonalpol besitzt Tupaia belangeri im Untersuchungszeitraum drei weitere Orte hohen Apoptoseaufkommens: (1) das chordoneurale Scharnier, das bei der Labormaus keine Apoptosehäufungen aufweist, (2) die Schwanzknospe und (3) das ventrale Mesoderm . Schließlich belegen unsere Rekonstruktionen erstmals, dass die Chorda dorsalis nicht nur von einer, sondern sogar von zwei dicht aufeinander folgenden Apoptosewellen in kraniokaudaler Richtung durchlaufen wird.de

dc.description.abstractengThe present work investigates, whether and to what extent apoptosis and proliferation contribute to the morphogenesis of somites in 12 to 16-day-old embryos of the primate-related Tupaia belangeri. To provide optimal conditions for structural analysis, semithin sections were studied using our previously established scanning and reconstruction system. We cannot confirm earlier reports which favoured a decisive role of cell death in the segmentation of the paraxial mesoderm underlying somite formation. In Tupaia belangeri, development of the intersomitic gaps starts independently in ventromedial and dorsolateral parts of the paraxial mesoderm. First description of intersomitic gap formation from two opposite sites may help to reconcile existing scientific controversies on this subject. Finally, we have identified candidates for "border cells" which may assist in intersomitic gap formation, and which have been postulated to exist based on quail-chicken transplantation experiments. Having available refined three-dimensional reconstruction techniques, it was possible to describe the epithelial-to-mesenchymal transition of somite cells into sclerotome cells in more detail than previously obtainable. Sclerotome formation starts medially, then spreads cranially and caudally in this medial position, and extends laterally with a delay. Confirming earlier findings in chicken and mouse embryos, accumulations of apoptotic cells were observed in the lateral subdomain of the sclerotome. Newly found simultaneous accumulations of apoptotic and proliferating cells in the ventral subdomain may contribute to the development of the perichordal sheath. In the central subdomain of the sclerotome which resides between the neural tube and the myotome, high proliferative activity occurs in the almost absence of apoptosis. In Tupaia belangeri, precursor cells of the epaxial myotome originate from the dorsomedial lip of the dermomyotome, whereas precursor cells of the hypaxial myotome develop from the ventrolateral lip. In the trunk, "de-epithelialization" of the dermomyotome starts centrally. In segments which lack hypaxial myotomes, onset of the de-epithelialization was noted in the ventrolateral lip. High proliferative activity among precursor cells of the myotomes was first observed in the dorsomedial and ventrolateral lips of the dermomyotome, thereafter spread to all four lips and, finally, was diffusely distributed in the entire dermomyotome. Large-scale apoptosis among presumed progenitor cells of the dermis was found in the dorsomedial lip as well as in central parts of the trunk dermomyotomes during de-epithelialization. Likewise, formation of hypaxial myotomes is preceded by massive apoptosis in the ventrolateral lip, whereas progenitor cells of the limb muscles are most probably subjected to apoptotic selection during migration toward their peripheral targets. Resembling all other studied vertebrates, the number of mitotic and apoptotic cells within the developing myotomes increases with time. Mutually exclusive proliferative maxima in dorsomedial and apoptotic maxima in ventrolateral parts of the myotomes appear to be another constant finding among representatives of all vertebrate classes. In Tupaia belangeri, simultaneous reconstructions of the paraxial and intermediate mesoderm, the lateral plate and the notochord revealed band-like accumulations of apoptotic cells which appeared to "separate" the lateral plate from all adjacent medial parts of the mesoderm. In addition, we postulate that pro-apoptotic factors released by the lateral plate may initiate large-scale apoptosis (1) in the lateral subdomain of the sclerotome, (2) in the ventrolateral lip of trunk dermomyotomes, (3) among migrating progenitor cells of the limb muscles which originate from the ventrolateral lip of the dermomyotome, and (4) in the ventrolateral half of the epaxial myotome. During the period of examination, embryos of Tupaia belangeri demonstrate three additional sites of massive apoptosis in the caudal pole: (1) the chordoneural hinge which lacks apoptotic maxima in mice, (2) the tail bud, and (3) the "ventral mesoderm". Finally, our reconstructions show for the first time that not one but two successive waves of apoptosis pass through the notochord in a craniocaudal direction.de
