This dissertation examines signalling of hematopoietic growth factors in hippocampal neurons and cortical astrocytes. The activity patterns of Akt, ERK1/2 and Stat3 after treatment with thrombopoietin (TPO) or granulocyte colony-stimulating factor (GCSF) were analyzed in different neural cell types with a focus on their relative contributions on both cell survival and proliferation. The actions of these growth factors showed remarkable cell type specificity. Also the importance of Stat5 for neuroprotective and neurotrophic actions of erythropoietin (EPO) was examined using neurons isolated from Stat5 knockout mice, and comparison to growth hormone. A model where neurotrophic but not neuroprotective effects of EPO depend on Stat5 signalling pathway is proposed.

Die vorliegende Dissertation untersucht die Signalgebung hämatopoietischer Wachstumsfaktoren in hippokampalen Neuronen sowie kortikalen Astrozyten. Analysiert werden im Einzelnen die Aktivitätsmuster von Akt, ERK1/2 und Stat3 nach Behandlung mit Thrombopoietin (TPO) oder Granulozyten-Kolonie-stimulierendem Faktor (GCSF) in verschieden Typen von Neuralzellen im Hinblick auf deren relative Beiträge sowohl zum zellulären Überleben, als auch zur Proliferation. Die Wirkung dieser Wachstumsfaktoren zeigte bemerkenswerte Zelltypspezifität. Des Weiteren wurde die Bedeutung von Stat5 für die neuroprotektive und neurotrophe Wirkungen von Erythropoietin (EPO) mit dem Einsatz isolierter Neuronen von Stat5- Knockout- Mäusen und im Vergleich zum Wachstumshormon erforscht. Ein Modell, in dem neurotrophe, aber nicht neuroprotektive Effekte von EPO von Stat5-Signalwegen abhängen, ist denkbar.
