dc.description.abstractSchwach leitende Kalzium-aktivierte Kaliumkanäle (SK-Kanäle) sind kalziumselektiv und voltunabhängige Kanäle, die durch intrazelluläres Kalzium während eines Aktionspotenzials aktiviert werden und eine wichtige Rolle in erregbaren Zellen spielen (Kohler, M. et al., 1996). In dieser Arbeit wurden sechs neue Spleißvarianten des Maus-SK2-Kanals durch Bioinformatik-Analyse gefunden, die darauf hinweisen, das SK2 auf post-transkriptioneller Ebene hoch dosiert ist. Zwei dieser Varianten, SK2P und SK2sh, sind artübergreifend stark erhalten und scheinen eine potenziell neue Funktion innerhalb der SK-Kanal-Familie zu haben. Die SK2CaMBD-Variante hatte ein unfertiges Stop-Kodon, die SK2R-Variante änderte nicht die SK2std bezügliche Protein-kodierende Region und die SK2-N-Terminus-Variante wurde nur in Herz- und Lebergewebe gefunden. Post-transkriptionelle Spaltung innerhalb eines drei Aminosäuren-Einsatzes am C-Endpunkt von SK2P führt zu einem 10KDa-Peptid, das einen SNARE-Bereich hat, der in die SNARE-Komplexbildung eingreifen könnte und modulierte schließlich Exozytose und Glutamat-Freisetzung. SK2P ist besonders in der CA3-Region des Maus-Hippokampus lokalisiert und ist nach Furchtkonditionierungstraining erhöht. Ein 10 KDA-Peptid, das höchstwahrscheinlich von der C-terminalen Region von SK2P abgespalten ist, wurde zu verschiedenen Zeitpunkten nach dem Training in Hippocampus-Fraktionen ermittelt. Da das 10-KDa-Peptid nicht im Hippocampus von unbehandelten Tieren nachgewiesen wurde, kann eine Rolle in der Gedächtnis-Konsolidierung angenommen werden. Die SK2sh-Variante, aus der drei mebranübergreifende Bereiche herausgeschnitten sind (insgesamt 140 Aminosäuren), scheint eine nichtkanalformierende Untereinheit zu sein und befindet sich in Hirnregionen, die reich an Astrozyten und Mikroglia sind. Da SK2P eine potenzielle Rolle in Glutamat-Freisetzung und Gedächtniskonsolidierung und SK2sh eine Rolle in der Glioneurotransmitter-Freisetzung eine Rolle spielen könnte, könnten Funktionen dieser Varianten die Ursache für die scheinbar widersprüchlichen Rollen von SK2-Kanälen in hippokampalem LTP und kontextuellem Furchtgedächtnis sein. Diese Arbeit zeigt auch, dass SK2P nach Stress stark hoch dosiert ist und die Hauptvariante für die stattfindende hohe Modulation in den kumulativen Auswirkungen von SK2 mRNA ist.. Alternatives Spleißen der SK2-primären Transkription fügt den SK2-Kanal-Funktionen im ZNS daher unerwartete Komplexität hinzu.de

dc.description.abstractengSmall conductance calcium-activated potassium channels (SK channels) are potassium selective and voltage independent channels, which are activated by intracellular calcium during an action potential and play an important role in excitable cells (Kohler, M. et al., 1996). In this study six novel splice variants of mouse SK2 channel were found through bioinformatics analysis, indicating that SK2 is highly regulated at the post transcriptional level. Two of these variants, SK2P and SK2sh, were highly conserved across species and seem to have potentially novel function within the SK channel family. The SK2CaMBD variant had a premature stop codon, the SK2R variant did not alter the protein-coding region relative to SK2std and the SK2-N-terminus variant was found only in heart and liver tissues. Post translational cleavage within a three amino acid insert at the C-terminus of SK2P results in a 10 KDa peptide, which has a SNARE domain that may interfere with the SNARE complex formation, ultimately modulating exocytosis and glutamate release. SK2P is specifically localized in the CA3 region of the mouse hippocampus and is up-regulated after fear conditioning training. A 10 KDa peptide, which is most likely cleaved off at the C-terminal region of SK2P was found at several time points after training in hippocampal fractions. Since the 10 KDa peptide was not detected in the hippocampus of naïve animals, a role in memory consolidation can be expected. The SK2sh variant, which has three transmembrane domains spliced out (a total of 140 amino acid), appears to be a non-channel forming subunit and is localized in regions rich in astrocytes and microglial in the brain. Since SK2P plays a potential role in glutamate release and memory consolidation and SK2sh might play a role in glioneurotransmitter release, functions of these variants could be the reason for the seemingly contradictory roles of SK2 channels in hippocampal LTP and contextual fear memory. This study also showed that SK2P is highly up-regulated after stress and is the major variant undergoing high modulation in the cumulative effects of SK2 mRNA. Alternative splicing of the SK2 primary transcript thus adds unexpected complexity to SK2 channel functions in the CNS.de
