dc.description.abstractDie Auflösung konventioneller Mikroskope ist begrenzt durch das Abbesche Beugungslimit. Die Auflösungsgrenze lässt sich jedoch fundamental brechen durch sequenzielles Schalten von Markierungsstoffen in der zu betrachtenden Probe zwischen einem detektierbaren An-Zustand und einem nicht detektierbaren Aus-Zustand. Dies kann zum Beispiel durch Fotoschalten geschehen, d.h. durch Einstrahlen von Licht geeigneter Wellenlänge. Die beiden derzeitigen Methoden, die das Schalten der Farbstoffe in dieser Form ausnutzen sind einerseits die RESOLFT-Mikroskopie (REversibleSaturableOpticaL Fluorescence Transitions) und die SMS-Mikroskopie (Single Marker Switching). In der vorliegenden Arbeit werden für beide Methoden neue Ansätze des Fotoschaltens von organischen Farbstoffen untersucht und die Ergebnisse dieser neuen Varianten der hochauflösenden Mikroskopie vorgestellt.de

dc.description.abstractengThe resolution of conventional microscopes is limited by Abbe"s diffraction barrier. The resolution barrier, however, can be fundamentally broken by sequentially switching marker substances in the sample between a detecable on-state and a non-detecable off-state. This can for example be achieved by photoswitching, i.e. by illumination by light of suitable wavelengths. The two current methods which utilise switching of the markers in this form are RESOLFT-microscopy (REversibleSaturableOpticaL Fluorescence Transitions) on the one hand and SMS-microscopy (Single Marker Switching) on the other hand. In this work, new approaches for the photoswitching of organic fluorophores for both methods are investigated and the results of these new variants of high-resolution microscopy are presented.de
