Fluorescence Microscopy Methods are widely used in the Life Sciences to visualize and spectroscopically analyze biologically processes. Single molecule methods, such as FCS (Fluorescence Correlation Spectroscopy) and SMT (Single Molecule Tracking), allow the investigation of single macromolecular complexes with high temporal resolution. Due to the non invasive nature of light microscopy those investigation can be done in living cells, something which is e.g. not possible with electron microscopy. Unfortunately light microscopy is limited in its spatial resolution by diffraction (> 200nm). Recently several approaches (e.g. STED, PALM, STORM) have been developed to overcome the resolution limit by taking advantage of the photophysical properties of the fluorescent dyes in use. The combination of single molecule methods and STED microscopy permits the investigation of biological questions simultaneously with a high temporal and spatial resolution. In the first part of this work I have characterized the physical properties of such a combination of a STED microscope and the FCS method and in the second part I could successfully show how this combination allows new insights into the dynamics of lipids within the cell membrane.

Fluoreszenzmikroskopische Verfahren werden in den Biowissenschaften routinemäßig zur Visualisierung und zur spektroskopischen Analyse von zellulären Abläufen verwendet. Spektroskopische Einzelmolekülmethoden, wie beispielsweise FCS (Fluorescence Correlation Spectroscopy) oder SMT (Single Molecule Tracking), erlauben es die Dynamik biologischer Prozesse auf der Ebene einzelner makromolekularer Komplexe mit hoher zeitlicher Auflösung zu betrachten. Aufgrund der nicht invasiven Natur der lichtmikroskopie ist es hierbei möglich die Untersuchungen, im Gegensatz zu elektronenmikroskopischen Verfahren, direkt in lebenden Zellen durchzuführen. Jedoch, durch das Beugungslimit optischer Verfahren bedingt, nicht mit hoher räumlicher Auflösung (>200 nm). In jüngster Zeit wurden einige hochauflösende Mikroskopieverfahren (z.B. STED, PALM, STORM) entwickelt, die diese Beschränkung der Fluoreszenzmikroskopie durch Nutzung photophysikalischer Eigenschaften der Fluoreszenzfarbstoffe umgehen. Die Kombination Spektroskopische Einzelmolekülmethoden mit hochauflösender STED - Mikroskopie ermöglicht es biologische Fragestellungen simultan mit hoher räumlicher und hoher zeitlicher Auflösung zu untersuchen. Im ersten Teil meiner Arbeit charakterisiere ich die physikalischen Eigenschaften einer solchen Kombination von STED Mikroskop und FCS Methode und im zweiten Teil zeige ich wie man mit Hilfe dieser Methoden neue Einblicke in das dynamische Verhalten der Lipide in der Zellmembran gewinnen kann.
