dc.description.abstractEndocytose reguliert eine Vielfalt von zellulären Prozessen, einschließlich die Proteinkomposition der Plasmamembrane, interzelluläre Signalkaskaden und die Zellpolarität. Die Zellpolarität wird von asymmetrisch an die Plasmamembrane lokalisierten Proteinkomplexen reguliert. Defekte in der Endocytose können zu einer fehlerhaften Lokalisation von Polaritätsproteinen und schließlich zu einem Verlust der Zellpolarität führen. Die kleine GTPase Cdc42 reguliert die Endocytose von Polaritätsproteinen in Drosophila Epithelzellen. Während die Rolle von Cdc42 in der Regulation von frühen Endosomen relativ gut beschrieben ist, ist bisher nicht bekannt, wie Cdc42 die späteren Schritte der Endocytose beeinflusst. In dieser Arbeit habe ich ein bisher unbekanntes und nicht charakterisiertes Protein Rush hour (Rush) beschrieben. Rush hour ist hochkonserviert und reguliert in Interaktion mit Cdc42 die Bildung von späten Endosomen. Rush bindet an Lipide und lagert sich an die laterale Plasmamembrane von Epithelzellen und an späte Endosomen an. Rush reguliert die Bildung der späten Endosomen und rush-Mutanten zeigen eine reduzierte Größe der späten Endosomen. Zusammen mit Cdc42 reguliert Rush die Endocytose der apikalen Polaritätsproteins Crumbs. Der Verlust von rush rettet den Überexpressionsphänotyp von dominant-negativem Cdc42. Rush bindet direkt an Rab GDI und könnte die Aktivierung der Rab Proteine an die Membranen der späten Endosomen beeinflussen. Zellen, die Rush überprimieren, zeigen eine verminderte Färbung mit dem Lysosommaker Lysotracker. Dies deutet auf die mögliche Rolle von Rush im Übergang von späten Endosomen zu Lysosomen hin. Außerdem ruft Rush das Clustering der vergrößerten frühen Endosomen, die durch Überexpression von konstitutiv aktivem Rab5 enstanden sind, hervor. Diese Ergebnisse deuten auf eine mögliche Funktion von Rush in der Regulation des Übergangs sowohl zwischen frühen und späten Endosomen, als auch zwischen späten Endosomen und Lysosomen.de

dc.description.abstractengEndocytosis regulates multiple cellular processes, including the protein composition of the plasma membrane, intercellular signaling and cell polarity. Cell polarity is defined by asymmetric localization of membrane-associated protein complexes. Defects in endocytosis can lead to faulty distribution of polarity proteins and ultimately to the loss of cell polarity. The small GTPase Cdc42 has recently been described to regulate endocytosis of polarity proteins in Drosophila epithelial cells. While the role of Cdc42 in the regulation of early endocytotic steps is quite well described, it is still not known how Cdc42 influences late endosome formation. Here I describe Rush hour (Rush), a previously uncharacterized and highly conserved protein that genetically interacts with Cdc42 in regulation of late endosome formation. Rush is a lipid interacting protein and localizes to the lateral plasma membrane of epithelial cells and to late endosomes. Rush promotes late endosome formation and rush mutants have a decreased late endosome size. Rush interacts with Cdc42 in regulation of endocytosis of the apical polarity protein Crumbs, and loss of rush rescued the phenotype caused by overexpression of dominant negative Cdc42. Rush binds directly to Rab GDI and could regulate endocytosis by affecting Rab activation on the membrane of late endosomes. Lysosomal marker staining is decreased in Rush overexpressing cells, indicating that Rush might regulate the transition between late endosomes and lysosomes. In addition, Rush causes fractionation of enlarged early endosomes that are formed by overexpression of constitutively active Rab5. These results suggest that Rush might act in trafficking steps both from the early to late endosomes and from late endosomes to lysosomes.de
