dc.description.abstractMitglieder der Unc-13/Munc-13-Protein-Familie sind Komponenten der präsynaptischen aktiven Zone und essentielle für das Priming synaptischer Vesikel bei der regulierten Exozytose. Unc-13/Munc-13-Proteine vermitteln das Priming synaptischer Vesikel, indem sie die offene Konformation des Q-SNAREs Syntaxin stabilisieren und so die Bildung des SNARE-Komplexes begünstigen. Die Unc-13/Munc-13-Protein-Familie ist evolutionär hoch konserviert und Vergleiche von Unc-13-Homologen verschiedener Species zeigen, dass Munc13-Gene aus zwei Modulen bestehen, die unabhängig evolvierten und unabhängige Funktionen haben. Die C-terminalen zwei Drittel der Proteine sind hoch konserviert zwischen Drosophila, C. elegans, Maus, Ratte und Mensch. Von den bekannten Unc-13-Homologen besitzen nur Munc13-1 und ubMunc13-2 einen N-terminus mit Homologie zur L-Region von Unc-13 aus C. elegans. RIM1alpha ist eine Komponente der aktiven Zone, das an den konservierten N-terminus von Munc13-1 und ubMunc13-2 bindet und ebenfalls am Priming synaptischer Vesikel beteiligt ist. In der vorliegenden Studie wurden mit Hilfe eines Mutagenesescreens im Yeast-Two-Hybrid -System in Kombination mit biochemischen und zellbiologischen Testverfahren punktmutante Varianten von Munc13-1 und ubMunc13-2 identifiziert, die nicht mehr an RIM1alpha binden. Mit Hilfe dieser Mutanten und RIM1alpha-defizienten Mäusen wurde gezeigt, dass die RIM1alpha-Bidung von Munc13-1 und ubMunc13-2 deren Verankerung an der aktiven Zone vermittelt.de

dc.description.abstractengMembers of the Unc-13/Munc13 protein family are components of the presynaptic active zone and essential vesicle priming factors in regulated exocytosis. They are thought to mediate vesicle priming by stabilizing the open conformation of the Q-SNARE Syntaxin, thereby promoting the SNARE core complex assembly. The Unc-13/Munc13 protein family is evolutionarily highly conserved and comparison of the various Unc-13 homologs in different species demonstrates that the respective genes consist of two modules that have evolved differentially and may have independent functional roles. The carboxy-terminal two-thirds of the protein are highly conserved between Drosophila, C. elegans, mouse, rat, and human, whereas of the known mammalian Unc-13 homologs, only Munc13-1 and ubMunc13-2 contain an amino-terminus with homology to the Unc-13 L region of C. elegans. RIM is an active zone protein, that has been found to bind this conserved amino-terminus of Munc13-1 and ubMunc13-2 and has been! proposed to act at multiple steps of the synaptic vesicle cycle, including targeting/docking, priming and fusion of synaptic vesicles. In the present study, we used a random mutagenesis approach in combination with yeast two-hybrid technology, biochemical assays as well as immunocytochemistry to identify residue(s) in the Munc13-1/ubMunc13-2 sequence that are critical for RIM binding. The ultimate aim of these studies is to develop the basis for subtle genetic interference with the RIM/Munc13-interaction in mice, which will allow a detailed functional characterization of this synaptic regulatory protein-protein interaction.de
