dc.description.abstract | Im Rahmen der Abwehrreaktion der Pflanze gegen
Pathogene steigen die Mengen endogener Salizylsäure an,
wodurch eine Reihe von Abwehrgenen aktiviert wird. Im
Rahmen dieser Arbeit wurden zwei Mutanten, aca41 und aca38#8, charakterisiert, die
verschiedene Abwehrgene konstitutiv exprimieren. Trotz
der konstitutiven Expression der Abwehrgene zeigte die
Mutante aca41 im Vergleich
zu Kontrollpflanzen nach Infektion mit virulenten
Bakterienstämmen nur eine leicht erhöhte Resistenz.
Durch Makroarrayanalysen wurden in aca41 drei konstitutiv exprimierte Gene
identifiziert, die normalerweise durch oxidativen
Stress induziert werden und deren Expression eine
Resistenz gegen oxidativen Stress vermittelt.
Wachstumsversuche auf Paraquat, einer Superoxid
erzeugenden Substanz, zeigten, dass aca41 eine erhöhte Toleranz gegenüber
oxidativen Stress aufweist. Obwohl in aca38#8 dieselben oxidativen Stressgene
wie in aca41 konstitutiv
exprimiert wurden, führte dies nicht zu einer erhöhten
Resistenz von aca38#8
gegenüber oxidativen Stressbedingungen. Weiterhin
zeigten sowohl aca41 als
auch aca38#8 einen
geringeren Wasserverbrauch als 5GuDe. Durch Messung der
Transpirationsrate, konnte für Linie aca41 gezeigt werden, dass sie unter
Wassermangelbedingungen ihre Stomata schneller schließt
und dass ihre minimale Transpirationsrate (Verdunstung
über die Cuticula) im Vergleich zur Kontrollpflanze
verringert war. Durch Kreuzung der Linie aca41 mit der NahG-Pflanze, wurde die Abhängigkeit
der verschiedenen Phänotypen vom Salizylsäuregehalt in
der Pflanze untersucht. Da NahG für eine Salizylat-Hydroxylase
codiert, die Salizylsäure in Catechol umwandelt, können
in diesen Pflanzen keine erhöhten Salizylsäuremengen
akkumulieren. Es konnte gezeigt werden, dass in
aca41 die Expression der
PR-Gene Salizylsäure
abhängig, aber sowohl die Expression der oxidativen
Stressgene als auch die erhöhte Austrocknungstoleranz
Salizylsäure unabhängig war. | de |

dc.description.abstracteng | One of the main components of the plant defense
response during an infection is the raise of endogenous
salicylic acid levels which leads to an increased
expression of defense genes. In this work two mutant
plants, aca41 and
aca38#8, were characterized
which have a constitutive expression of different
defense genes. In spite of the constitutive defense
gene expression the resistance of aca41 against virulent bacteria is only
slightly higher than in control plants. Array
experiments with aca41
leads to the identification of three genes which were
expressed usually by oxidative stress and mediate
resistance to oxidative stress. Growth experiments with
aca41 on media containing
paraquat, a superoxid generating substance, show that
this mutant is more resistant to oxidative stress.
aca38#8 which expresses the
same oxidative stress genes as aca41 is not able to grow under this
stress conditions. Additionally the mutant plants
turned out to have a higher tolerance against water
shortage. Measurement of the transpiration rate of
aca41 showed that this
plant closes their stomata faster by drought and that
the evaporation of the cuticula is lower than in
control plants. Analysis of the aca41 phenotype in the nahG background showed that the PR gene
expression is dependend of the endogenous salicylic
acid level while the expression of the oxidative stress
genes and the drought tolerance is independend. | de |
