dc.description.abstract | Eine Vergleichsanalyse strukturverwandter Indolamine
von Melatonin zeigte, dass bereits kleine Änderungen
der Substituenten erhebliche Konsequenzen in der
Oxidationschemie nach sich ziehen.Analysiert wurden hierbei die Substitutionen von
Hydroxy- oder Methoxygruppen an C-5 bzw. C-6 und einem
aliphatischen N-Acetylrest. Im Einzelnen wurden
Tryptamin, 5-Methoxytryptamin, N-Acetyltryptamin,
Serotonin, N-Acetylserotonin, Melatonin und
6-Methoxymelatonin in folgenden Oxidationssystemen
verglichen: Photooxidation mit Protoporphyrin-IX,
Hämin-katalysiertes H2O2-System und Elektronendonation
an ABTSKationradikalen (ABTS*+).5-Methoxytryptamin und N-Acetylserotonin zeigten
einen beschleunigten Umsatz im
Protoporphyrin-IX-System. Im ABTS*+-System hingegen ist
5-Methoxytryptamin langsamer als Melatonin. Im
Vergleich zu N-Acetyltryptamin wurde N-Acetylserotonin
schneller oxidiert. Die Reaktion von Serotonin verlief
langsamer als die von Tryptamin. 5-Methoxytryptamin und
Tryptamin ergaben eine erheblich größere Zahl an
Oxidationsprodukten als Melatonin.Der Sonderfall des nicht natürlichen
6-Methoxymelatonin zeigte, dass die Einführung der
zusätzlichen Methoxygruppe an C6 zu einer
Reaktivitätssteigerung und damit schnellerem Umsatz
führte, dies bereits in Hydroxylradikal-unabhängigen
Reaktionen. Im Produktspektrum konnten zusätzliche
Substanzen detektiert werden, deren nähere Untersuchung
noch aussteht. Diese Ergebnisse einer abweichenden
Oxidationschemie lassen einen therapeutischen Einsatz
von 6-Methoxymelatonin, zum jetzigen Zeitpunkt, ohne
Kenntnis der zusätzlichen Produkte, noch nicht
angeraten erscheinen. Das wichtigste Resultat der hier
vorgenommen Produktanalyse führt zu einer Erweiterung
der Radikalfängerkaskade von Melatonin.Zuvor war angenommen worden, dass diese mit der
Bildung von N1-Acetyl-N2-formyl-5- methoxykynuramin
(AFMK) endet. Es wird nunmehr gezeigt, dass AFMK durch
radikalische Mechanismen zu Substanzen umgesetzt wird,
die einer bislang unbekannten Klasse von
Melatonin-Metaboliten angehören. Sie entstehen
ebenfalls beim Umsatz von Melatonin als
Ausgangsprodukt. Hierbei handelt es sich um
C2-substituierte 3-Indolinone, welche jede für sich
eine chemisch neue Verbindung darstellt. Eine
stöchiometrische Verbrauchsanalyse in
ABTS*+-Reduktionssystem zeigt, dass bis zu den neuen
Produkten insgesamt 10 Radikale eliminiert werden.
Zuvor war nur ein Verbrauch von 4 Radikalen bis zum
AFMK bekannt gewesen.Bei den in diesem Prozess neu entstanden Produkten
handelt es sich um N-(1-Formyl-5-
methoxy-3-oxo-2,3-dihydro-1H-indol-2-ylidenmethyl)-acetamid
[Produkt 2] und N-(1-
Formyl-2-hydroxy-5-methoxy-3-oxo-2,3-dihydro-1H-indol-2-ylmethyl)-acetamid
[Produkt 3]. Die neuen Substanzen können als ein Art
von Rezyklisierungsprodukte des AFMK angesehen werden
und bilden zum Teil E,Z-Isomere [Produkt 2a/b]. Bei
zwei weiteren Verbindungen [Produkt 4 und Produkt 5]
handelt es sich nach Daten der Massenspektroskopie
offenbar um deformylierte Analoge. Im
ABTS*+-Reaktionssystem ergab sich das Produkt 3 aus dem
Racemat der Produkte 2a/b, jedoch nicht umgekehrt. Die
Ergebnisse dieser Arbeit unterstreichen die
außergewöhnlichen Redox-Eigenschaften von Melatonin
einschließlich der aus ihm hervorgehenden
Reaktionsprodukte. | de |

dc.description.abstracteng | A comparison analysis in structure related
Indolamine of Melatonin showed that small changes of
Substituenten already draw considerable losses after
themselves in oxidation chemistry. Analysed were the
substitutions of Hydroxy or Methoxygroups at C 5 or C 6
and an aliphatic N acetylene rests. Tryptamin,
5-Methoxytryptamin, N-Acetyltryptamin, Serotonin,
N-Acetylserotonin, Melatonin and 6-Methoxymelatonin
were compared in the following oxidation systems:Photo oxidation with Protoporphyrin-IX, Hemin
catalyzed H2O2 system and electron donation at ABTS
cation radicals (ABTS ). 5-Methoxytryptamin and N
Acetylserotonin showed a faster reaction in the
Protoporphyrin-IX system. In ABTS-system the reaction
of 5-Methoxytryptamin is slower than Melatonin. In
comparison with N Acetyltryptamin, N Acetylserotonin
was oxidized faster. The reaction of Serotonin was
slower than the reaction of Tryptamin.
5-Methoxytryptamin and Tryptamin yielded a
substantially larger number of oxidation products than
Melatonin. The special case of the not natural
substance 6-methoxymelatonin shows that an additional
methoxy group at C6 leads to a higher activity, already
in hydroxyl radical independent reactions. Additional
substances could be seen in the product line whose
nearer examination is still due. These results of a
divergent oxidation chemistry make a therapeutical use,
at the present time, of 6-Methoxymelatonin not
recommended without knowledge of the additional
products. The most important result of this product
analysis leads out to an expansion of the radical
scavenging cascade of melatonin. It had been assumed
before, that the radical scavenging ends with (N1
Acetyl-N2 formyl 5-methoxykynuramine) (AFMK). Now it
could be shown that AFMK is converted by radical
mechanisms to substances which belong to a till now
unknown class of melatonin metabolites.They were also produced when instead of AFMK
melatonin were used. These C2-substituted 3-Indolinones
are each for them self new chemical substances. A
stoichiometric consumption analysis in ABTS reduced
assay shows that altogether 10 radicals are eliminated
up to the new products. Only a consumption of 4
radicals had been known up to the AFMK before. In this
process the new products are N-(1-Formyl 5-methoxy
3-oxo 2,3-dihydro 1 H-indol 2-ylidenmethyl) acetamid
[product 2] and N-(1-Formyl 2-hydroxy 5-methoxy 3-oxo
2,3-dihydro 1 H-indol 2-ylmethyl) acetamid [product 3].
The new substances can be seen as re-cyclization
produces of AFMK which are partly E, Z isomers [product
2 a/b]. Another two compounds [product 4 and product 5]
are apparently deformylated analogous according to data
of mass spectrometry. In ABTS-reaction system , the
racemat of product 2 a/b gives products 3, but not the
reverse. The results of this reseach underline the
unusual Redox mechanism of melatonin including the
reaction products resulting from it. | de |
