dc.description.abstractDer Adaptor-Protein Komplex 1 (AP-1) vermittelt die Bildung von transportvesikeln am trans-Golgi Netzwerk und ist Bestandteil Clathrin umhüllter transportvesikel. AP-1 vermittelt die Clathrin Bindung an Vesikel und bindet zudem an Sequenzmotive in zytoplasmatischen Domänen von transmembranproteinen. Seine Funktion wird in der Sortierung von Proteinen in Clathrin-transportvesikel gesehen. Der AP-1 Komplex besteht aus 4 Untereinheiten, deren Funktionen für die Proteinsortierung und die Vesikelbildung nur in Teilen verstanden sind. Die beiden grossen Untereinheiten γ1 und β1 bestehen jeweils aus einer N-terminalen globulären Domäne und einer C-terminalen globulären Domäne, die eine Vielzahl von Proteinen binden, deren Funktionen bei der Vesikelbildung nicht verstanden sind. Die μ1A Untereinheit des Komplexes ist essentiel für die Membranbindung des AP-1 und bindet transmembranproteine, die durch den AP-1 Komplex transportiert werden. Mit Ausnahme des β 1 Adaptin existieren von allen Untereinheiten des AP-1 Isoformen, von denen die Mehrzahl gewebsspezifisch expremiert werden. Die Funktionsanalyse von μ2/μ1A-Adaptin Chimären zeigte, dass der Bereich geringster Sequenzhomologie eine Bedeutung für die AP-1 vermittelte Proteinsortierung hat. Die Membranbindung des AP-1 Komplexes mit dem μ2/μ1A Chimären ist stabiler. Mittels des Y2H Verfahrens wurde ein bislang nicht charakterisiertes Protein identifiziert, das mit hoher Affinität und Spezifität an diese Domäne bindet. Seine Überexpression führt zu einer verminderten Membranbindung des AP-1, nicht des chimären Komplexes. Vom σ1-Adaptin existieren drei Isoformen σ1A, σB und σC. Das σ1B transskript wird zudem gewebsspezifisch alternativ gespleisst. Es sollten spezifische Funktionen für σ1 Isoformen identifiziert werden. Da ein γ1 Adaptin Bindemotif in der Isoform σ1B-3 existiert, nicht aber in den anderen, wurde dieses Motif analysiert. Es interagierte nicht mit γ1, jedoch lies sich durch Änderung einer einzigen flankierenden Aminosäure, eine Bindung erreichen.Es wurde die Sortierung des Sortilin Rezeptors in μ1A-sowie σ1B Adaptin-defizienten Zellen untersucht. In μ1A -/- zellen ist Sortilin von seiner normalen trans-Golgi Netzwerk Lokalisation in frühe Endosomen umverteilt. Daraufhin wurde die Bindung zweier Sortierungsmotife des Sortilin überprüft. Das Tyrosin-basierende Motif zeigte eine Interaktion mit dem μ1A Adaptin. Das di-Leucin-basierende Motif zeigte die höchste Affinität zu γ1/σ1B Hemikomplexen. Embryonale Fibroblasten einer σ1B "knock-out" Maus zeigten jedoch keine Sortilinfehlsortierung. Die Interaktion des Sortilins mit μ1A Adaptin scheint daher für eine Sortierung zwischen dem trans-Golgi Netzwerk und Endosomen in diesen Zellen ausreichend zu sein. Adipocyten von σ1B-defizienten Mäusen hatten jedoch eine vierfach höhere Menge an Sortilin. σ1B-Adaptin vermittelt daher in Adipocyten die Sortierung entlang eines gewebsspezifischen transportweges.de

dc.description.abstractengThe Adaptor-Protein complex (AP-1) mediates the formation of transport vesicles at the trans-Golgi network and is a component of Clathrin Coated Vesicles. Besides facilitating the binding of clathrin to vesicles, AP-1 also binds the sorting signal motif present in the cytoplasmic domains of transmembrane proteins and thereby functions to sort proteins into transport vesicles. The AP-1 complex consists of 4 subunits, whose functions in the protein sorting and the vesicle formation are only partially understood. Both the large subunits γ1 and β1 consist of, a N-terminal globular domain and a C-terminal globular domain, which bind a μltiplicity of proteins, whose functions in vesicle formation are not known. The μ1A subunit of the complex is essential for the membrane binding of the AP-1 and binds transmembrane proteins, which are transported by the AP-1 complex. With the exception of β1-adaptin, isoforms exist from all subunits of the AP-1, which show tissue specific expression. The functional analysis of a μ2/μ1A-adaptin chimera showed that the domain of μ1A, exhibiting the least sequence homology, has a role in the AP-1 mediated protein sorting. The membrane binding of the AP-1 complex containing the μ2/μ1A chimera, is more stable. Using Y2H method, an uncharacterized protein was identified, which binds with high affinity and specificity to this domain. Its over-expression leads to a decreased membrane binding of the AP-1, but not of the chimeric complex. Three isoforms are found for the σ1-adaptin viz., σ1A, σB and σC, whose functions are not known. Further, the σ1B transcript shows tissue specific alternative splicing. Since a γ1-adaptin binding motif is present in the isoform σ1B-3, this motif was analyzed. It did not interact with γ1, however, interaction was observed by changing only one flanking aminoacid.The sorting of the Sortilin receptor was examined in μ1A-deficient and also in σ1B-adaptin deficient cells. In μ1A -/- cells, sortilin is mislocalized from its normal trans-Golgi network localization to early Endosomes. Thereupon, the role of two sorting motifs of sortilin was examined. The Tyrosine-based Motif showed an interaction with the μ1A-adaptin. The di-Leucine based Motif showed the highest affinity for γ1/σ1B hemi-complex. However, embryonic fibroblasts of σ1B "knock out" mouse showed no mis-sorting of sortilin. The interaction of sortilin with μ1A-adaptin, therefore seems to be sufficient for sorting between the trans-Golgi network and endosomes in these cells. Adipocytes of σ1B deficient mice had however a fourfold higher quantity of sortilin. In adipocytes, the σ1B-adaptin might be involved in a tissue specific transport pathway.de
