dc.description.abstract | Das Proepikard ist eine embryonale
Vorläuferzellpopulation, die die Zellen des Epikards,
die Hauptmasse des kardialen Interstitiums sowie
sämtliche Zellen des koronaren Blutgefäßsystems
liefert. Das Proepikard besteht aus Mesothel- und
Mesenchymzellen, die sich im Bereich des venösen Pols
des embryonalen Herzschlauchs bilden. Die vorliegende
Arbeit umfasst vier Teilstudien, in denen Aspekte der
Proepikardinduktion und differenzierung untersucht
wurden. In der ersten Teilstudie wurde die Entwicklung
des Proepikards im Hühner- und Mausembryo dokumentiert.
Es wurde gezeigt, dass sich paarige Proepikardanlagen
im Hühnerembryo in einem bilateral asymmetrischen und
im Mausembryo in einem bilateral symmetrischen Muster
entwickeln und dass die beiden Proepikardanlagen von
Hühnerembryonen unterschiedliche Lagebeziehungen zu
benachbarten Geweben wie dem Dottersackmesoderm und dem
Kammermyokard der Herzschleife besitzen. Basierend auf
diesen Befunden wurde postuliert, dass diese Gewebe
einen hemmenden Einfluss auf die linke bzw. einen
induzierenden Einfluss auf die rechte Proepikardanlage
haben könnten. In der zweiten Teilstudie wurden die
Wirkungen von Dottermesoderm und Kammermyokard auf die
Proepikardentwicklung experimentell getestet. Hierbei
konnte ein Einfluss dieser Gewebe auf die
Proepikardentwicklung ausgeschlossen werden. Basierend
auf diesem Befund wird postuliert, dass die
asymmetrische Entwicklung des Proepikards im
Hühnerembryo durch rechts/links-seitenspezifische
Signale gesteuert wird. Das Ziel der dritten Teilstudie
war die Entwicklung experimenteller Verfahren zur
Testung verschiedener Faktoren auf ihre mögliche
Proepikard induzierende bzw. Proepikard hemmende
Wirkung. Aufgrund von technischen Problemen, deren
Ursache in der speziellen Morphologie und Physiologie
der Proepikardregion lagen, konnte leider kein
entsprechendes experimentelles Verfahren entwickelt
werden. In der vierten Teilstudie wurde untersucht, ob
das kardiale Lymphgefäßsystem vom Proepikard abstammt.
Durch immunhistochemische Untersuchungen von Whole
mount-Präparaten mit dem Lymphendothelmarker Prox1
konnte die Entwicklung des kardialen Lymphgefäßsystems
dokumentiert werden. Weiter wurden
Wachtel/Huhn-Chimären konstruiert, bei denen ein
Wachtelproepikard in einen Hühnerembryo transplantiert
wurde. Nach Reinkubation und Doppelfärbung mit dem
Wachtelendothel spezifischen Antikörper QH1 und dem
Lymphendothelmarker Prox1 konnte eine Beteiligung der
proepikardialen Vorläuferzellen an der Bildung des
kardialen Lymphgefäßendothels ausgeschlossen

dc.description.abstracteng | The proepicardium is an embryonic precursor cell
population, which delivers the epicardium, the cardiac
interstitium and all cells of the coronary blood
vessels. The proepicardium consists of mesothelial and
mesenchymal cells and forms near to the venous pole of
the embryonic heart tube. In four substudies the
aspects of the induction and differentiation of the
proepicardial development was examined. In the first
substudy, the development of the proepicardium was
documented in the chicken and mouse embryo. It was
found that paired proepicardial anlagen developed in a
bilaterally asymmetric pattern in chicken embryo and in
a bilaterally symmetric pattern in mouse embryo.
Further it was found that both proepicardial anlagen in
chicken embryo had different relationships to
neighboring tissues such as the yolk sac and the
ventricular myocardium. Based on these findings it was
postulated that these tissues might have repressing and
inducing influences on the proepicardial anlagen. In
the second substudy the effects of the yolk sac
mesoderm and the ventricular myocardium on the
development of the proepicardium was experimentally
tested. Influences by these tissues could be excluded.
Based on these findings it is postulated that the
asymmetric development of the proepicardium in chicken
embryo is regulated by left/right signaling. The goal
of the third substudy was to develop an experimental
procedure for testing different factors that could
possibly have an inducing or repressing effect on the
proepicardium. Due to technical problems which were
caused by the special morphology and physiology of the
proepicardial region, no appropriate experimental
procedure could be established. In the fourth substudy
it was examined whether the cardiac lymphatic
endothelial vessels are derived from the proepicardium.
The development of the cardiac lymphatic vessels could
be documented by immunohistochemical examinations of
whole mount specimen with the lymphendothelial marker
Prox1. Further quail/chicken chimeras were constructed
by transplanting a quail proepicardium into a chicken
embryo. After reincubation and double staining with the
quail endothelium specific antibody QH1 and the
lymphendothelial marker Prox1 a participation of
proepicardial precursor cells in the development of the
cardiac lymphatic vessels could be excluded. | de |
