dc.description.abstract | Adrenalen Steroidhormonen (z.B. Cortisol) fällt eine
zentrale Schlüsselstellung in der Regulation und
Erhaltung der metabolischen Homeostase zu. Die
Sekretion der Glucokortikoide durch die
adrenokortikalen Zellen in das Blut ist kaum erforscht.
Basierend auf der lipophilen Struktur der
Steroidhormone wurde lange Zeit postuliert, das die
Sekretion durch einfache Diffusion vonstatten geht. Vor
kurzem konnte jedoch gezeigt werden, das die Freigabe
von Cortisol aus - sowie die Aufnahme von radioaktiv
markiertem PAH in - adrenokortikale Zellen des Rindes
ähnliche physiologische Charakteristik wie der renale
organische Anionentauscher OAT1 zeigen. In dieser
Arbeit sollte daher der Frage nachgegangen werden, ob
der organische Anionentransporter OAT1 möglicherweise
an der Cortisolsekretion durch die humane Nebennieren-
Zelllinie NCI-H295R beteiligt ist. Die basale
Cortisolsekretion über 24h konnte durch die
Vorbehandlung mit ACTH um das dreifache gesteigert
werden, durch Vorhandlung mit Forskolin sogar um den
Faktor 30. Eine Inkubation mit PAH über 24h inhibierte
teilweise die Cortisol Abgabe, wohingegen eine Hemmung
der Sekretion durch Cimetidine relativ ausgeprägt
nachgewiesen werden konnte, was auf Unterschiede
zwischen NCI-H295R Zellen und den Zellen der Nebenniere
des Rindes hindeutet. Mittels RT-PCR konnte keine
Expression von humanem OAT1 und OAT2 sowohl in
NCI-H295R als auch in normalem humanen
Nebennierengewebe nachgewiesen werden, für OAT3 und
OAT4 konnte jedoch mRNA detektiert werden. In Studien
mit HEK-293 Zellen, die stabil mit humanem OAT1, OAT3
und OAT4 transfiziert wurden, zeigte sich eine
schwächere Interaktion von Cortisol mit hOAT1 und hOAT4
als mit hOAT3. Werden humanes OAT1, OAT3 und OAT4 in
Xenopus laevis Oozyten exprimiert, so zeigt (verglichen
mit nicht exprimierenden Kontroll-Oozyten) nur hOAT3
eine Aufnahme von radioaktiv markiertem Cortisol.Die Cortisolaufnahme konnte mittels eines Kt Wertes
von 2.5 µM gesättigt werden. Die Aufnahme von
radioaktiv markiertem Östronsulfat (rl-Östronsulfat.)
wurde durch unmarkiertes Cortisol mit einer IC50 von
15.6 µM inhibiert. Die Experimente mit NCI-H295R Zellen
zeigten eine Sättigung der Aufnahme von
rl-Östronsulfat, einem wichtigen Subtrat von hOAT3
(Ki=9.8µM). Die Inhibition mittels unmarkiertem DHEAS
und Cortisol war durch IC50 Werte von 10.6µM (DHEAS)
und 38.9µM (Cortisol) möglich. Eine Reduktion der
Aufnahme von rl-Östronsulfat in NCI-H295R-Zellen konnte
durch potente Inhibitoren von hOAT3, z.B. Probenecid,
Cimetidin und Glutarat gezeigt werden. In NCI-H295R-
Zellen konnte durch vorheriges Beladen mit Glutarat
oder Cortisol eine Stimulation der Aufnahme von
rl-Östronsulfat erzielt werden. Ähnliche Beobachtungen
der Stimulation für die Aufnahme von PAH wurden durch
vorheriges Beladen mit PAH, Gluratat oder Cortisol
gemacht. In NCI-H295R Zellen konnte eine Aufnahme von
rl-Östronsulfat und PAH signifikant gesteigert werden,
indem die Zellen über einen Zeitraum von 24h mit
Forskolin inkubiert wurden. Über semiquantitative
RT-PCR wurde eine gesteigerte hOAT3 mRNA Expression
nach Inkubation mit Forskolin gezeigt. Ebenso
resultierte eine Inkubation mit Forskolin in einer
signifikant erhöhten mRNA Expression der Enzyme der
Steroid-Biosynthese (z.B. StAR, CYP17, 3ßHSD und
CYP21A2). Mittels Immunfluoreszenz wurde über einen
spezifischen hOAT3 Antikörper die Expression auf
NCI-H295R Zellen bestätigt. Diese Expression war durch
eine 24h Behandlung mit Forskolin steigerbar.Zusammenfassend lassen diese Daten vermuten das
hOAT3 funktionell in NCI-H295R Zellen exprimiert wird
und den Cortisol-Anionen Austausch ermöglicht. Neben
anderen Mechanismen ist nach den in dieser Arbeit
durchgeführten Analysen eine Beteiligung von OAT3 an
der Sekretion von Cortisol aus humanen
Nebennierenzellen wahrscheinlich. | de |

dc.description.abstracteng | Adrenal steroid hormones (e.g., cortisol) play a
pivotal role in the regulation and maintenance of
metabolic homeostasis. The secretion of glucocorticoids
from adrenocortical cells into the blood is poorly
understood. It has long been postulated that this
occurs via simple diffusion, based on the lipophilic
structure of steroid hormones. Previously, it has been
demonstrated that cortisol release from and uptake of
radiolabeled PAH into bovine adrenocortical cells
showed physiological characteristics similar to the
renal/organic anion exchanger OAT1. In this study we
investigated whether an organic anion transporter (OAT)
may play a role in cortisol release from the human
adrenal cell line, NCI-H295R. Basal 24 h cortisol
secretion increased up to threefold by pretreatment
with ACTH, and up to thirtyfold with forskolin.
Incubation for 24 h with PAH partially inhibited
cortisol release, while cimetidine inhibition was
relatively pronounced, indicating some differences
between NCI-H295R cells and bovine adrenal cells.
RT-PCR did not reveal an expression of human OAT1 and
OAT2, but OAT3 and OAT4 mRNA was detected in NCI-H295R
cells as well as in normal human adrenal tissue. The
studies with HEK-293 cells stably transfected with
human OAT1, OAT3, and OAT4 showed a low interaction of
cortisol with hOAT1 and hOAT4 as compared to hOAT3.
When human OAT1, OAT3, and OAT4 were expressed in
Xenopus laevis oocytes, only hOAT3 showed radiolabeled
cortisol uptake beyond non-expressing control oocytes.
Cortisol uptake was saturable with an apparent Kt value
of 2.4 µM, and radiolabeled estrone sulfate uptake was
inhibited by unlabeled cortisol with an IC50 of 15.6
µM. The experiments in NCI-H295R cells showed a
saturable radiolabeled estrone sulfate uptake, a potent
substrate of hOAT3, with a Ki value of 9.8 µM. The
inhibition with unlabeled DHEAS and cortisol resulted
in IC50 values of 10.6 and 38.9 µM, respectively. The
radiolabeled estrone sulfate uptake in NCI-H295R cells
was decreased by potent inhibitors of hOAT3, i.e.
probenecid, cimetidine, and glutarate. In NCI-H295R
cells, radiolabeled estrone sulfate uptake was
trans-stimulated by preloading with glutarate or
cortisol. Likewise, radiolabeled PAH uptake was
trans-stimulated by preloading the cells with PAH,
glutarate, or cortisol. The 24 h forskolin treatment
significantly increased radiolabeled estrone sulfate
and PAH uptake in NCI-H295R cells. Semi-quantitative
RT-PCR showed an increased hOAT3 mRNA expression after
pretreatment with forskolin. Forskolin-treatment
induced a significant increase in the enzymes of
steroids biosynthesis, i.e. StAR, CYP17, 3âHSD, and
CYP21A2. Immunolocalization studies for hOAT3 resulted
in expression of hOAT3 in NCI-H295R cells. There was a
high increase in OAT3 expression by a 24 h treatment
with forskolin. Our data suggests that OAT3 is
functionally expressed in NCI-H295R cells and is able
to perform cortisol/anion exchange. Thereby, OAT3 may -
among other release mechanisms - contribute to cortisol
efflux from human adrenal cells. | de |
