dc.description.abstract | Peptidantibiotika aus drei verschiedenen Familien
wurden durch röntgenkristallographische Methoden
untersucht. Es konnten bis atomarer Auflösung streuende
Kristalle aus den Peptaibol-Antibiotika Cephaibol A, B
und C erhalten werden, und die Raumstrukturen wurden
dementsprechend aufgeklärt. In allen drei Fallen nahmen
die Peptidketten eine helikale, am zentralen
Hydroxyprolin gebeugte Konformation an. Die
Überlagerung der Moleküle zeigt, dass die N-terminale
Helix starr, während die C-terminale wesentlich
flexibler ist. Obwohl die räumlichen Strukturen sich
nur geringfügig unterscheiden, ist die Anordnung von
Wasserstoffbrücken deutlich verschieden; nur beim
Cephaibol C konnte aus der Kristallpackung die Bildung
eines Membrankanals interpretiert werden, die für die
biologische Aktivität ein typisches Merkmal sein
soll.Die Kristallisation von Feglymycin, einem
einzigartigen Antibiotikum bezüglich seiner
Aminosäuresequenz und seinem antibakteriellen Spektrum,
führte zu zwei Kristallformen. In beiden Fallen bildet
das Peptid identische asymmetrische Dimere mit einer
antiparallelen, doppelsträngigen
Doppel-β9.0-Helix, die dem
Membrankanal-Peptid Gramicidin ähnelt. Der
Feglymycin-Kanal ist jedoch von zwei
Phenylalanin-Seitenketten gesperrt und möglicherweise
zu kurz, eine biologische Membran zu überspannen. Die
Dimerstruktur wird von zahlreichen Wasserstoffbrücken
stabilisiert und ist wahrscheinlich starr. Mit Sicht
auf die Struktur könnte Feglymycin eher als ein
unspezifischer Enzyminhibitor wirken denn als
Membrankanal.Friulimicin und Tsushimycin gehören zu
Lipopeptid-Antibiotika. Obwohl zwei Kristallformen von
Friulimicin erhalten wurden, konnte das Phasenproblem
in keinem Fall gelöst werden. Im Gegensatz dazu
ermöglichten die bis zu hoher Auflösung streuenden
Tsushimycin-Kristalle die Strukturlösung durch ab
initio direkte Methode. Die asymmetrische Einheit
enthält 12 Moleküle, die jeweils ein
Ca2+-Ion binden, das eine wichtige Rolle in
der Stabilisierung der Raumstruktur zu spielen scheint.
Die Moleküle bilden Dimere mittels zusätzlicher
Ca2+-Ionen. Die Kristallpackung eifert der
Struktur einer Micelle nach, indem die Moleküle polare
und apolare Hohlräume trennen. Der antibakterielle
Wirkungsmechanismus konnte aus der Struktur nicht
eindeutig festgestellt werden, aber vieles deutet
darauf hin, dass das Antibiotikum nicht in die
bakterielle Zelle gelangen kann, sondern an der
Oberfläche wirkt. | de |

dc.description.abstracteng | Peptide antibiotics belonging to three different
families were investigated using X-ray crystallography.
The peptaibol antibiotics cephaibol A, B and C gave
well-diffracting crystals and their structures were
determined at 0.9 Å resolution. They adopt a helical
conformation with a sharp bend at the central
hydroxyproline. Superposition of the models shows that
the N-terminal helix is rigid and the C-terminus is
flexible, but the differences in three-dimensional
structure are negligible. However, there are variations
in the hydrogen-bonding pattern for the three
structures, and only in the case of cephaibol C does
the packing emulate the formation of a membrane channel
believed to be important for their biological
function.Feglymycin, a unique antibiotic in terms of amino
acid sequence and antibacterial spectrum, yielded two
crystal modifications. In both cases the peptide was
found to form asymmetric dimers in form of an
antiparallel double-stranded double
β9.0-helix and resemble the membrane channel
peptide gramicidin. However, the feglymycin channel is
blocked by two phenylalanine sidechains and it is
probably too short to span a biological membrane. The
dimeric structure is stabilised by numerous hydrogen
bonds and therefore it is probably rigid. Based on the
structure, it is more likely that feglymycin is an
unspecific enzyme inhibitor than a membrane agent.Friulimicin and tsushimycin belong to the group of
lipopeptide antibiotics. Two crystal forms were
obtained from friulimicin, but in neither case could
the phase problem be solved. Tsushimycin gave
well-diffracting crystals and was solved using ab
initio direct methods. The structure contains 12
molecules; each of these binds a Ca2+-ion
that seems to be important in stabilising the
three-dimensional structure. The molecules associate to
dimers via interactions mediated by additional
Ca2+-ions. The crystal packing emulates the
structure of a micelle and the molecules act as
separators between polar and non-polar cavities. The
mechanism of antimicrobial action cannot be
unambiguously identified from the crystal structure,
but it seems likely that the molecules cannot enter the
bacterial cell and act on its surface. | de |
