dc.description.abstract | Kleine, nicht kodierende Mikro-RNAs regulieren die
Translation von vielen mRNAs durch unvollständige
Basen-Paarung in der 3 nicht-translatierten Region.
Drei konservierte Gene, welche drei miR-196 RNAs
kodieren, befinden sich in den Hoxa, Hoxb
beziehungsweise Hoxd Gengruppierungen von Säugern und
Vögeln. Aufgrund von Bioinformatik- und von
Reportergen-Studien wurden die drei Maus Gene Hoxa7,
b8, und c8 als Ziele von miR-196 identifiziert.
Überexpression von miR-196 beeinflusst die ektopische
Induktion von HOXB8 durch Retinsäure in den Anlagen der
Vordergliedmaße des Huhns. In Wirbeltier-Embryonen sind
Hox-Gene von der Gastrulation bis zur Organentwicklung
an der antero-posterioren Musterbildung beteiligt. In
dem sich entwickelnden Rückenmark sind sie
Determinanten der Motorneuronen-Identität und besonders
wichtig für ihre kolumnare Anordnung. Die gegenwärtige
Studie untersucht die Funktion von miR-196 in der
Entwicklung des lumbaren Rückenmarks.
Durch RNA In Situ-Hybridisierung und durch doppelte
Sensor-Tracer-Analyse wurde gezeigt, dass die
Expressions-Bereiche von miR-196 und HOXB8 entlang der
antero-posterioren-Achse des Rückenmarks sich
gegenseitig ausschliessen. HOXB8 war im Hals- und
Brustbereich des Neuralrohrs exprimiert, während
miR-196 angrenzend an die hintere Grenze des
HOXB8-Bereiches im lumbaren Bereich vorhanden war.
MiR-196 unterdrückte spezifisch die Translation von
HOXB8 durch Wechselwirkung mit seiner 3 UTR, sowohl in
kultivierten Zellen, als auch im lebenden Embryo.
Dieser Regulierungseffekt wurde zusätzlich durch
knockdown -Ansätze bestätigt, in denen HOXB8 von
dieser Repression befreit wurde. Die Störung des
Gleichgewichts zwischen HOXB8 und miR-196 lumbaren
Neuralrohr durch Misexpression von HOXB8 oder durch
Hemmung der miR196-Aktivität beeinflusste die zeitliche
Entwicklung der Motorneuronen. Die Anzahl der
Progenitor-Zellen war erhöht und ihr Austritt aus dem
Zellzyklus wurde verzögert,! so dass die Entwicklung
der postmitotischen Motorneuronen und Axonprojektion
beeinträchtigt war. Diese Daten deuten darauf hin, dass
HOXB8 im lumbaren Neuralrohr in der Entwicklungsphase
herunterreguliert werden muss, wenn die
Motorneuron-Vorläuferzellen aus dem Zellzyklus
austreten und Neuronen geboren werden sollen. MiR-196
wird zur richtigen Zeit in den richtigen Zellen
exprimiert, um die Abwesenheit von HOXB8-Protein durch
direktes Zusammenspiel mit HOXB8 mRNA zu
vermitteln.
Vorangegangene Studien hatten die Bedeutung der
anterioren Hox-Gen-Expressionsgrenzen betont, und einen
kombinatorische Mechanismus und /oder eine posteriore
Prevalenz diskutiert. Die gegenwärtige Studie liefert
Belege für eine mikroRNA vermittelte Definition der
posterioren Expressions-Grenze eines Hox-Gens, als
einen zusätzlichen Mechanismus zur Definition von
zellulären Identität entlang der Körperachse. | de |

dc.description.abstracteng | Small, non-coding microRNAs regulate the translation
of many mRNAs through imperfect base pairing in the 3
untranslated region. Three conserved genes, encoding
three miR-196 RNAs are located in the mammalian and
avian Hoxa, Hoxb and Hoxd gene clusters, respectively.
Based on bioinformatics and reporter gene studies, the
three murine genes Hoxa7, b8 and c8 were identified as
targets of miR-196. Overexpression of miR-196 impaired
the ectopic induction of HOXB8 by retinoic acid in the
chick forelimbs. Hox genes are involved in
antero-posterior patterning of vertebrate embryos, from
gastrulation to organogenesis. In the developing spinal
cord they are determinants of motor neurons identity,
and in particular important for their columnar
organisation. The current study investigates the
function of miR-196 in the development of the lumbar
spinal cord.
By RNA in situ hybridization and double sensor-tracer
analysis, it is shown here that miR-196 and HOXB8 adopt
mutually exclusive expression domains along the
antero-posterior axis of the chick spinal cord. HOXB8
is expressed in the cervical and thoracic region of the
neural tube, whereas miR-196 is expressed in the lumbar
region adjacent to the posterior boundary of the HOXB8
domain. miR-196 specifically repressed translation of
HOXB8 via interaction with its 3 UTR both in cultured
cells and in the living embryo. This regulatory effect
was further confirmed by knockdown approaches relieving
HOXB8 from repression. Interfering with the balance
between HOXB8 and miR-196 by the misexpression of HOXB8
in the lumbar neural tube, or by inhibition of miR-196
activity, affected the timing of motor neuron
development. The number of progenitor cells was
increased, and their exit from the cell cycle delayed,
so that the development of postmitotic motor neurons
and their axonal projections were impaired. These data
suggest a requirement for HOXB8 downregulation from the
lumbar neural tube in a developmental phase, when motor
neuron progenitors are supposed to exit the cell cycle
and neurons are born. miR-196 is expressed at the right
time in the right cells to mediate the absence of HOXB8
protein via a direct interaction with HOXB8 mRNA.
Previous studies had emphasized the importance of
anterior Hox gene expression boundaries, indicating a
combinatorial mechanism and/or a posterior prevalence.
The present study provides evidence for a microRNA
mediated definition of the posterior expression
boundary of a Hox gene, as an additional mechanism to
define cellular identities along the body axis. | de |
