dc.description.abstract | Satb1, ein spezielles AT-reiche Sequenz bindendes
Protein 1, ist ein Transkriptionsfaktor, der an der
Regulierung der gewebespezifischen Organisation des
Chromatins verantwortlich ist. Es ist in den Kernen
lokalisiert und umgibt das Heterochromatin, in dem es
eine käfigartige Netzwerkstruktur bildet. Außerdem
regelt es wahrscheinlich die Transkription durch
spezifische Bindung an BUR Sequenz im Bereich der
Erkennungsstelle der Matrix Anlage-Regionen (MAR) der
genomischen DNA.Satb2, ein Homolog des Satb1, wurde von Victors
Tarabykin´s Arbeitsgruppe durch die Methode der
Subtraktionshybridisierung identifiziert, die auf dem
Screening von Genen basiert, die für die Kontrolle der
neuronalen Differenzierung verantwortlich sind. Wir
haben festgestellt, dass Satb2 ausschließlich in einer
Subpopulation von postmitotischen Neuronen der oberen
Schichten (II-IV) der kortikalen Platte exprimiert
wird, während Satb1 hauptsächlich in den unteren
Schichten (V-VI) und in den Schichten I-II exprimiert
ist.Zur Charakterisierung der zwei Subpopulationen von
Zellen, haben wir eine Mauslinie generiert, in der die
Satb1 kodierende Sequenz in den Satb2 Gen-Locus
integriert wurde, so dass den Mutanten nicht nur Satb2
fehlt, sondern zusätzlich Satb1 in den Zellen mit
transkriptional aktivem Satb2 Promotor ektopische
überexprimiert ist.Wir haben bereits gezeigt, dass der vollständige
Knockout von Satb2 zu verschiedenen craniofaciale
Abnormalitäten und Fehlbildungen in der Hirnrinde
führt. In dem Satb1- Knockin findet man auch einige
Probleme in der Lamination und Migration der Neuronen
vor, jedoch zeigen sie keine starke craniofaciale
Verformung, die zu postnatalem Tod führt, wie es im
Satb2 Knockout der Fall ist.Die ektopische Überexpression von Satb1 unter dem
Satb2 Promotor scheint weder die axonalen Verbindungen
in der Hirnrinde noch die Defekte in der Lamination
vollständig wieder kompensieren zu können und führt
möglicherweise zu einer erhöhten Heterodimerisation von
Satb1 und Satb2, was eine Veränderung der Ziel-
Spezifität von Satb2 im Ctip2 Locus zur Folge hat. | de |

dc.description.abstracteng | Satb1, special AT-rich sequence binding protein 1,
is a transcription factor that is involved in the
regulation of tissue-specific organization of
chromatin. It is localized at nuclei and surrounds
heterochromatin, forming a cage-like network structure
and seems to regulate transcription by specifically
binding to BUR sequence in recognition region of matrix
attachment regions (MAR) of genomic DNA.Satb2, a close homologue of Satb1, was identified by
Victor Tarabykin s group in a subtraction hybridization
based screen of genes controlling neural
differentiation. We found Satb2 to be expressed
exclusively in a subpopulation of postmitotic neurons
of the upper layers (II- IV) of the cortical plate,
whereas Satb1 is expressed mostly in deep layers (V-VI)
and in layer I-II.In order to characterize the two subpopulations of
cells, we have generated a mouse line where the Satb1
coding sequence has been knocked into the Satb2 gene
locus, such that the mutant not only lacks Satb2, but
also ectopically overexpresses Satb1 in cells with
transcriptionally active Satb2 promoter. We have
already shown that total knockout of Satb2 leads to
several craniofacial abnormalities and malformations in
the cerebral cortex. Satb1 k.i also present some
lamination as well as neuronal migration problems but
they do not show a strong craniofacial deformation that
leads to postnatal dead as in Satb2 k.o .The ectopic overexpression of Satb1 under the Satb2
promoter seems not completely restore axonal
connectivity in the cortex nor the lamination defects
and possibly leads to an increase in heterodimerisation
of Satb1 and Satb2, leading to a change in target
specificity of Satb2 at the Ctip2 locus. | de |
