Macroautophagy is a starvation-induced protein degradation pathway that nonselectively delivers cytosolic material and organelles to the vacuole (lysosome) for degradation. The Cvt-pathway (Cytoplasm to vacuole targeting) is one of the selective subtypes of autophagy. This pathway acts constitutively under nutrient-rich conditions and selectively transports specific proteins to the vacuole. Each subtype is dependent on a specific set of Atg-proteins along with associated proteins. Atg9 physically interacts with Atg2. The Split-Ubiquitin system was used to look for further interacting partners of Atg9. Mostly proteins of the 40S and 60S ribosomal subunits were found as putative partners. It has been shown that both the 40S and 60S are specifically degraded through a pathway termed ribophagy. Interactions with Atg9 and ribosomal proteins in this screen are probably unspecific. Another protein that was screened is the Cvt-specific protein Atg21. Beside a variety of ribosomal proteins Egd2 was found as a putative interaction partner. Deletion of EGD2 does not lead to a maturation-defect of proApe1 during the Cvt-pathway and Atg21 is not mislocalised. Further experiments showed that egd2Δ-cells also do not show a defect in vacuole fragmentation or inheritance. Atg18, Atg21 and Ygr223c are homologous proteins that have distinct functions in autophagy. This study shows that vacuole fragmentation in response to hyperosmotic shock is strongly reduced in atg18Δ-cells. Reduction in the atg21Δ-strain is not as severe and ygr223cΔ shows wildtype phenotype. All three deletions have a slightly reduced inheritance frequency compared to the wildtype. The three homologous proteins also share the phospatidylinositol-phosphate (PIP) binding motif FRRG. Replacing the FRRG motif with FTTG in Atg18 and Atg21, results in their mislocalization. The FRRG motif in Atg18 is not essential for vacuolar fission but Atg18FTTG showed a significant decrease in the fragmentation rate comparable to that of the atg18Δ-strain. Overexpression of GFP-YGR223C leads to the secretion of carboxypeptidase Y (CPY). This lead to the assumption that Ygr223c might bind and titrate an unknown binding partner needed for the functional CPY-transport. Simultaneous overexpression of Ygr223c and this partner should restore proper CPY-sorting. Co-overexpression of Ygr223c and a gene bank in a CPY-Overlay-Assay did not lead to the identification of an interacting partner for Ygr223c. Trs85, a non-essential component of the TRAPP I and II complex, is needed for the Cvt-pathway and for full activity during autophagy. The function of Trs85 during the Cvt-pathway could therefore involve a specific TRAPP complex. Experiments to isolate this putative complex were carried out with the TAP (tandem affinity purification)-method. Using Bet3-TAP as a control, lead to the almost complete isolation of the TRAPP II complex. Experiments with Trs85-TAP did not result in the identification of new binding partners for Trs85. Induction of ER stress through incubation of cells with either tunicamycin (TM) oder dithitreitol (DTT), leads to the induction of ER-phagy. It was shown in this study that many Atg- and autophagy-associated proteins are needed for viabilty under ER-stress conditions and a lot of deletion strains show a significant reduction in cell growth. It was further shown that ER-phagy might not be classical microautophagy since GFP-Atg8 and Pgk1-GFP are not degraded under ER-stress conditions. This highlighted the idea to look for a specific marker protein to monitor ER-phagy. The tested candidates Kar2, Sec63 and Sec61 were no suitable markers.

Makroautophagie ist ein Prozess der eukaryotischen
Organismen unter anderem das Überleben bei
Nährstoffmangel sichert. Für Autophagie sind selektive
Subtypen bekannt. Einer ist der Cvt (Cytoplasm to
vacuole targeting)-Weg. Dieser ist unter
Wachstumsbedingungen aktiv und transportiert
spezifische Proteine unter Ausschluss von Zytosol in
Cvt Vesikeln zur Vakuole. Für ihre Funktion benötigen
alle Subtypen eine spezifische Auswahl von Autophagie-
und weiteren assoziierten Proteinen. Mit Hilfe des
Split-Ubiquitin System wurde in dieser Arbeit nach
Interaktionspartnern für die Proteine Atg9 und das
Atg21 gesucht. Mögliche Interaktionen mit Atg9 wurden
vor allem mit Bestandteilen der ribosomalen 40S und 60S
Untereinheit gefunden. Diese werden in der selektiven
Ribophagie abgebaut. Unspezifische Interaktionen in
diesem System scheinen auf Grund der räumlichen Nähe
der Proteine bei diesem Prozess möglich. Für Atg21
wurde mit dem Split-Ubiquitin System neben vielen
ribosomalen Proteinen auch Egd2 als möglicher
Interaktionspartner entdeckt. Egd2Δ-Zellen zeigen
jedoch keinen Cvt-Defekt. Die Lokalisierung von Atg21
ist in diesen Zellen ebenfalls nicht beeinträchtigt.
Zudem zeigen egd2Δ-Zellen keinen Defekt bei der
Vakuolenfragmentierung oder vererbung. Die drei
sequenzhomologen Proteine Atg18, Atg21 und Ygr223c
haben unterschiedliche Funktionen in autophagischen
Prozessen. In dieser Arbeit wurde gezeigt, dass die
Vakuolenfragmentierung, als Antwort auf
hyperosmotischen Stress, in atg18Δ-Zellen signifikant
und atg21Δ-Zellen deutlich gestört ist. Ygr223cΔ-Zellen
zeigen hingegen den Wildtypphänotyp. Die Vererbung der
Vakuole in knospenden Zellen ist dagegen in allen drei
Deletionsstämmen reduziert. Den drei Proteinen gemein
ist das Phospatidylinositol-Phosphat (PIP)-Bindemotiv
FRRG. Die Mutation zu FTTG in Atg18 und Atg21 führt zur
fehlerhaften Lokalisierung in der Zelle. Zusätzlich
verliert Atg18 die Fähigkeit an PI3P und PI(3,5)P2 zu
binden. Dieses Motiv ist auch wichtig für die Fähigkeit
zur Vakuole nfragmentierung. Atg18FTTG-Zellen zeigen
den gleichen Phänotyp wie atg18Δ-Zellen. Die
Überexpression von GFP-YGR223C bewirkt die Sekretion
von CPY. Dies führte zur Annahme, dass überschüssiges
Ygr223c einen unbekannten Faktor binden könnte, der für
den CPY-Transport benötigt wird. Die gleichzeitige
Überexpression dieses vermuteten Bindungspartners
sollte den Defekt wieder aufheben. In einem
CPY-Overlay-Assay mit Hilfe einer
Überexpressions-Genbank konnte kein solcher Faktor
identifiziert werden. Trs85 ist ein nicht-essentieller
Bestandteil der TRAPP (transport protein
particle)-Komplexe. In trs85Δ-Zellen ist der Cvt-Weg
blockiert und Autophagie um 50% reduziert. Mit Hilfe
der TAP (tandem affinity purification)-Methode sollte
für Trs85 nach Bindungspartnern gesucht werden, um zu
klären, ob Trs85 Bestandteil eines Cvt-spezifischen
TRAPP Komplexes ist. Mit Bet3-TAP (als Kontrolle)
konnte der TRAPP II Komplex fast vollständig isoliert
werden. Für Trs58 konnten keine neuen
Interaktionspartner identifiziert werden. Induzierter
ER-Stress durch Inkubation von Zellen in Tunicamycin
(TM) oder Dithitreitol (DTT) führt zur Induktion von
ER-Phagie. In dieser Arbeit wurde gezeigt, dass viele
zusätzliche Atg-Proteine und Autophagie-assozierte
Proteine essentiell für das Wachstum unter ER-Stress
sind. Weitere Deletionsstämme zeigten dazu eine
signifikante Reduktion im Wachstum. ER-Phagie scheint
keine echte Makroautophagie zu sein. Die Markerproteine
für Autophagie, GFP-Atg8 und Pgk1-GFP, wurden unter
ER-Stress nicht in der Vakuole abgebaut. Experimente
zur Identifizierung eines geeigneten Markerproteins für
die Analyse der ER-Phagie zeigten, dass Kar2, Sec63 und
Sec61 keine geeigneten Markerproteine sind.
