dc.description.abstract | Das Gram-negative Bodenbakterium Acinetobacter sp. BD413 zeichnet sich
durch eine hohe Kompetenz für die Aufnahme freier DNA
aus und besitzt die Fähigkeit auf feuchten Oberflächen
zu gleiten. Genetische und biochemische Untersuchungen
zeigen eine Beteiligung von Kompetenzproteinen, die
Ähnlichkeiten zu Biosynthesefaktoren von Typ-IV-Pili
aufweisen, an der natürlichen Transformation; diese
übernehmen aber keine essentielle Funktion bei der
Fortbewegung des Bakteriums. Nach diesen Ergebnissen
lautete die zentrale Frage, ob die beiden Pilustypen
(dicke und dünne Pili) von Acinetobacter sp. BD413 für die
DNA-Aufnahme oder die gleitende Fortbewegung essentiell
sind. Während nach genetischen und physiologischen
Studien dieser vorliegenden Arbeit sich die dünnen Pili
zur Klasse der Typ-I-Pili zuordnen lassen, weisen die
flexiblen dicken Pili keine Verwandtschaft zu bereits
charakterisierten filamentösen Strukturen auf. Da
sowohl die natürliche Transformation als auch die
gleitende Fortbewegung bei Pilusmutanten nicht
beeinträchtigt ist, sind die Pilusstrukturen von BD413
nicht an diesen beiden physiologischen Prozessen
beteiligt.Elektronenmikroskopische Untersuchungen an den
dicken Pili demonstrieren weiterhin einen für
filamentöse Strukturen charakteristischen helikalen
Anordnung der 28 kDa Untereinheiten. Die variable
Steighöhe der Helix von 4 bis 10 nm begründet
vermutlich die hohe Biegsamkeit dieser Struktur. Im
Gegensatz dazu werden die 16 kDa Untereinheiten des
dünnen Pilus in linearer Weise assembliert.Weitere Untersuchungen betreffen das
DNA-Aufnahmesystem, welches den Transport der freien
DNA durch die Zellhülle erlaubt. Hierbei stellte sich
heraus, daß der Typ-IV-Pilin-ähnliche Kompetenzfaktor
ComP keine Untereinheit der Pili von BD413 darstellt,
aber das Bakterium nach Überexpression von comP einen weiteren Pilustyp
synthetisiert. | de |

dc.description.abstracteng | The Gram negative soil bacterium Acinetobacter sp. BD413 is known for
its high competence of natural transformation and its
movement on solid surfaces. Genetic and biochemical
studies of the DNA transformation apparatus indicated
homologues of type IV pili subunits being involved in
natural transformation of strain BD413 but not in its
gliding motility. The central question arising from
these findings is whether BD413 thin or thick pili are
involved in DNA translocation or movement. For this
investigation it was necessary to characterize the
pilus structures.Genetical and physiological observations of this
work demonstrate the relatedness of BD413 bundle
forming thin pili to the type I family (chaperone/usher
pathway). The 28 kDa subunit of the flexible thick pili
show no similarity to known pilus structure proteins.
Unaffected competence for natural transformation and
unaffected gliding motility of pilus mutant strains
indicate that thin and thick pilus structures are not
involved in DNA uptake and movement on solid surfaces.
Electron microscopic studies demonstrate that the thick
pilus subunit structure follows the common composition
of filaments in assembling a stable helical structure.
The 6 nm pilus possesses a flexible height per turn of
4-10 nm that might be explain the flexibility of the
filament. In contrast to this, the 16 kDa subunits of
the straight 2-3 nm thin pili are assembled into a
linear manner.Furthermore, our interest focuses on the DNA-uptake
system that mediates free DNA to passage through the
outer and inner membrane. Molecular analyses of the
transformation machinery revealed that the competence
factor ComP a type IV pilin-like protein is not
assembled into thin or thick pili of BD413.
Interestingly, after over-expression comP the bacterium synthesises another
pilus structures on the cell surface. | de |
