dc.description.abstract | Das Wind-Protein ist ein Schlüsselfaktor/ wichtiger
Faktor in der Dorsoventral-Entwicklung während der
Embryogenese in Drosophila. Es wurde gezeigt, dass Wind
essentiell ist für die Translokation des Proteins Pipe.
Der Mechanismus bleibt aber weiterhin unklar. In dieser
Arbeit zeigen wir die 3D Struktur von Wind durch
Kristallographie bei einer Auflösung von 1,9 Ả. Durch
die Auflösung der Struktur und darauf basierende
site-directed-Mutagenese-Studien können wir unseren
Einblick bezüglich Struktur und Funktion von Wind
vertiefen.Wind wird der PDI-verwandten ER-Proteine zugeordnet
und setzt sich aus 2 unterschiedlichen Domänen
zusammen: einer N-terminalen Thioredoxin-ähnlichen
b-Domäne und einer C-terminalen helikalen D-Domäne
zusammen. Beide sind über eine Linker-Sequenz
miteinander verbunden. Aufgrund der Linker-Flexibilität
kann die Orientierung der 2 Domänen variieren. Dieses
könnte für die Substrat-Bindung wichtig sein.Wind liegt als Homodimer vor, sowohl in 3D-Struktur
als auch in Lösung. Dieses Dimer wird durch eine
Kopf-Schwanz -Anordnung der b-Domänen bebildet, ohne
jegliche Beteiligung der D-Domänen. Bei der
Dimerisierung bildet sich eine tiefe Spalte, welche
gross genug ist, um kleine peptide zu binden/
festzuhalten. Diese Spalte ist negativ geladen und
weist beträchtliche Hydrophobizität auf der Oberfläche
auf. Die Aminosäurereste an der Oberfläche in der Nähe
der Spalte sind in der PDI-Dß-Familie konserviert.
Obwohl die b-Domäne mit einer möglichen
Substrat-Bindestelle nah des Dimerspalts essentiell
ist, spielt aber auch die D-Domäne für die Funktion des
Proteins Wind eine wichtige Rolle. | de |

dc.description.abstracteng | Wind is a key factor to trigger dorsal-ventral
patterning in the Drosophila embryo development.
Genetic experiments indicate that Wind is required for
targeting the dorsal-ventral patterning protein, Pipe,
from the ER to the Golgi. So far the mechanism is
unknown at the molecular level. Here, we have
determined the three-dimensional structure of Wind with
X-ray crystallography to 1.9 Å resolution. This crystal
structure and the site directed mutagenesis studies
based on it greatly promote our insight into the
relationship of the structure and function of Wind.Wind is a PDI-related ER resident protein consisting
of two distinct domains: an N-terminal thioredoxin-like
b-domain and a C-terminal helical D-domain, connected
by a short linker. Due to the flexibility of the
linker, the orientation of the two domains could vary,
which might be important for substrate capture.Wind exists as a homodimer both in the crystal and
in solution. The dimer is formed by a head-to-tail
arrangement of the b-domains without the participation
of the D-domains. The dimerization yields a deep dimer
cleft, which is large enough to hold a small peptide.
This dimer cleft is negatively charged, with a
considerable hydrophobic band spanning the top. And the
surface residues in the bank areas of this cleft are
rather conserved in PDI-Dβ family. Although the
b-domain is essential, in which a substrate binding
site is proposed near the dimer cleft, the D-domain is
also required for the function of Wind. | de |
