dc.description.abstract | Ziel der vorliegenden Arbeit war es, cytosolische
ABA-bindende Proteine aus Pflanzenextrakten zu
isolieren und nachzuweisen. Dazu wurden Proteine aus
Spinat- und Arabidopsis-Pflanzen durch verschiedene
proteinbiochemische Methoden wie Ammoniumsulfat-Fällung
und Säulenchromatographie aufgereinigt. Für den
Nachweis ABA-bindender Proteine wurde als
Detektionsverfahren ein nicht-kompetitiver ELISA
etabliert.
ABA-BSA-Konjugate wurden im ELISA als Sonden zum
Nachweis ABA-bindender Proteine eingesetzt. Dazu wurde
die Kopplung von ABA an BSA als Trägerprotein in dieser
Arbeit durchgeführt. Es konnte gezeigt werden, daß pro
BSA-Molekül durchschnittlich 21 ABA-Moleküle gebunden
wurden. Diese ABA-BSA-Konjugate wurden zur
Immunisierung von zwei Kaninchen verwendet. Die
gewonnenen polyklonalen Antikörper erwiesen sich als
spezifisch gegenüber ABA. Aus Experimenten, bei denen
die Konkurrenz von 3H-ABA und ABA um die Bindung an den
Antikörper untersucht wurde, konnte ein Kd-Wert von
36,8 nM für den ABA/Antikörper-Komplex erhalten
werden.
Eine Aufreinigung von Proteinen aus Spinatpflanzen
mittels Gelfiltrations-Chromatographie ergab keine
spezifische ABA-BSA-Konjugat Bindung. Von den durch
Aufreinigung von Arabidopsis-Proteinen mittels
Kationenaustauscher-Chromatographie erhaltenen
Fraktionen zeigten mehrere eine hohe
ABA-BSA-Konjugat-Bindung. Bei der weiteren Aufreinigung
einer dieser Fraktionen durch
ABA-Affinitäts-Chromatographie wurden zwei ABA-bindende
Proteine mit einem Molekulargewicht von 50 kDa und 52
kDa nachgewiesen. | de |

dc.description.abstracteng | Goal of this work was to isolate abscisic acid
binding proteins of spinach and Arabidopsis plants.
Therefore the use of different proteinbiochemic methods
like ammoniumsulfate-precipitation and chromatography
principles were necessary. For the detection of
abscisic acid binding proteins a none competitive ELISA
was estabilsh.
ABA-BSA-conjugates were used as linkers for the
detection of ABA-binding proteins. Therefore a coppling
reaction of ABA on BSA was estabilsh. One BSA-molecule
binds about 21 ABA-moleclues. The immunisation with
these conjugates of two rabbits leds to specific
antibodys against ABA. The Kd-constant was 36.8 nM,
measured with 3H-ABA.
Spinach proteins isolated by gel chromatography showed
no spezific binding of ABA-BSA-conjugates. The
protein-extracts of Arabidopsis separated by ion
exchange chromatography led to high ABA-BSA-conjugate
binding in some fractions. One of these fraction war
purified with ABA-affinity-chromatography. In the end
two Proteins with an molecular weight of 50 and 52 kDa
