dc.description.abstract | Evolutive Methoden der Molekularbiologie spielen bei
der Entwicklung von in Hinblick auf Funktion,
Stabilität oder Interaktion optimierten Biomolekülen
eine wichtige Rolle. Ein zentraler Aspekt ist dabei die
Bereitstellung von Proteinvarianten in mikrobiellen
Produzenten und deren Durchmusterung nach der
gewünschten Eigenschaft. In dieser Arbeit wurde ein
Verfahren entwickelt, das es erlaubt, Proteinvarianten
durch Kopplung an das Membranprotein EaeA (Intimin)
enterohämorrhagischer Escherichia
coli auf der Oberfläche von E. coli Zellen bereitzustellen und
damit einer Funktionsabfrage direkt zugänglich zu
machen. Verschiedene, sich hinsichtlich Größe, Struktur
und Stabilität unterscheidende Passagierproteine (u.a.
humanes Interleukin 4, Calmodulin, unterschiedliche
Inhibitorproteine) wurden in Bezug auf Effizienz der
Translokation durch die äußere Membran und deren
Abhängigkeit vom Zusatz chemischer Agenzien
(Reduktionsmittel, Detergenzien) untersucht. Aus diesen
Daten wurde ein Modell für die Intimin-vermittelten
Translokationen von Proteinen durch die äußere Membran
von Escherichia coli Zellen
abgeleitet. Die biotechnologische Anwendung der
Intimin-vermittelten Präsentation von
Peptid-/Proteinbibliotheken auf der bakteriellen
Zelloberfläche wurde in einem experimentellen Beispiel
demonstriert. | de |

dc.description.abstracteng | Methods of molecular evolution play an important
role for the optimization of biomolecules with respect
to function, stability, or interaction. A key aspect is
the generation of protein variants in microbial hosts
and their screening for the desired characteristics. In
this thesis a method for the display of peptides and
proteins on the surface of Escherichia coli bacteria via linkage
to the outer membrane protein EaeA (intimin) from
enterohemorrhagic Escherichia
coli is presented which allows for the efficient
screening of large libraries for a desired property.
Passenger domains of different size, molecular
structure, folding stability, and function (human
interleukin 4, calmodulin, different protein
inhibitors) were investigated with respect to
translocation competence across the bacterial outer
membrane and the influence of chemical compounds
(reducing agents, detergents) on their cell surface
exposition. Based on these data, a model for the
mechanism of protein translocation across the
Escherichia coli outer
membrane via intimin was deduced. Furthermore, it was
shown that molecular repertoires of peptides and
proteins can be stably maintained in E. coli and screened for specific
interaction partners by intimin bacterial surface
display and high-throughput fluorescence-activated
cell-sorting (FACS) using directly or indirectly
labelled target proteins. | de |
