dc.description.abstract | Die mitochondriale Atmungskette besteht
aus vier Membranproteinkomplexen für den Elektronentransport
(Komplex I-IV) und der ATP synthetisierenden F1F0ATPase. Diese
Komplexe bilden in der inneren mitochondrialen Membrane
Superkomplexes, so genannte Respirasomen. Trotz intensiver
Bemühungen seit vielen Jahren die Biogenese und Funktion dieser
Strukturen zu verstehen ist, sind viele Fragen noch ungeklärt. Das
Ziel dieser Arbeit war es neue Erkenntnisse zur Respirasombildung
und deren Rolle für die Zelle zu gewinnen. Mit Hilfe von
Affinitätschromatographie wurden die Superkomplexe der Atmungskette
aus S. cerevisiae Mitochondrien gereinigt, und neue Faktoren
identifiziert und charakterisiert. Rcf1 und Rcf2 wurden als Komplex
IV Untereinheiten identifiziert. Während Rcf2 nur in Hefearten
gefunden wurde, ist Rcf1 eine konservierte Untereinheit mit zwei
humanen Homologen, RCF1a und RCF1b. Hefezellen ohne Rcf1 haben eine
reduzierte Cytochrom-c-Oxidase Aktivität und reduziertes Wachstum,
vor allem in Hypoxie. Beide Mutanten, rcf1∆ und rcf2∆, haben eine
erhöhte Produktion reaktiver Sauerstoffspezies. Rcf1 vermittelt die
Oligomerisierung der Atmungskettenkomplexe, im besonderen die
Oligomere bestehend aus einem Komplex III Dimer und zwei Komplex IV
Monomeren. Des Weiteren wird Rcf1 für die effektive Assemblierung
von Cox13 und Rcf2 in die Cytochrom-c-Oxidase benötigt. Die
Identifizierung von Rcf1 hat auch gezeigt, dass in Mitochondrien
eine heterogene Cytochrom-c-Oxidase Population vorliegt. Die
verschiedenen Cytochrom-c-Oxidase Formen unterscheiden sich in der
Zusammenstellung der Untereinheiten. Wenigstens zwei Populationen
können unterschieden werden: eine Cox13 enthaltende und eine ohne
Cox13. Der Verlust der Cox13-enthaltenden Form wurde in rcf1∆
Mitochondrien beobachtet. Beide Formen können in
Atmungskettensuperkomplexe integrieren und damit verschiedene
Superkomplexe bilden. | de |

dc.description.abstracteng | The mitochondrial respiratory chain
consists of four membrane-embedded electron transport complexes
(complexes I – IV) and the ATP synthesizing F1F0ATPase. Within the
inner mitochondrial membrane these complexes assemble into
supercomplexes, forming so-called respirasomes. Although
understanding the biogenesis and function of these higher
oligomeric structures has been the subject of intense research for
many years it still remains largely enigmatic. The aim of this work
was to obtain novel insights into respirasome formation and their
cellular relevance. Using an affinity chromatography based strategy
respiratory supercomplexes from yeast, S. cerevisiae mitochondria
were purified and novel factors identified and characterized. Rcf1
and Rcf2 are reported to be complex IV components. While Rcf2 is
present only among the yeast species, Rcf1 is a conserved subunit
and has two human homologs, RCF1a and RCF1b. Yeast cells lacking
Rcf1 are affected in cytochrome oxidase activity and display
reduced growth efficiency, especially pronounced during hypoxia.
Both mutants, rcf1∆ and rcf2∆, show increased ROS production. Rcf1
mediates oligomerization of respiratory complexes, specifically for
the formation of oligomers consisting of complex III dimer and two
complex IV monomers. Additionally, Rcf1 is necessary for efficient
assembly of subunits Cox13 and Rcf2 into the cytochrome oxidase
complex. The identification of Rcf1 revealed that mitochondria
possess a heterogenous cytochrome oxidase population. The various
forms of cytochrome oxidase complexes differ in their subunit
composition. At least two cytochrome oxidase subpopulations can be
distinguished, one containing Cox13 and one that lacks the Cox13
subunit. Selective loss of the Cox13-containing form was observed
in rcf1∆ mitochondria. Both forms are able to incorporate into
supercomplexes, thus generating distinct forms of
