dc.description.abstract | Die CD8+ T zellvermittelte,
nichtzytotoxische antivirale Aktivität (CNAR) ist mit der Kontrolle
der HIV-Infektion in Langzeitüberlebenden assoziiert. Unter anderem
wird diese Aktivität einem unbekannten sezernierten Protein
zugeordnet, das als löslicher antiviraler CD8 Zellfaktor (CAF)
bezeichnet wird. Um Kandidatengene zu identifizieren, die mit CAF
oder CNAR assoziiert sind, wurden „Microarray“-Daten von CD8+
Zellen mit und ohne CNAR ausgewertet. Aus mehr als 50 differentiell
regulierten Genen wurden 16 ausgewählt und mittels der qRT-PCR in
CD8+ Zellen von 21 Rhesusaffen (Macaca mulatta) untersucht. FAM26F
war das einzige Kandidatengen, das in CNAR-positiven und negativen
CD8+ Zellen von SIV-infizierten und nicht infizierten Affen
signifikant unterschiedlich exprimiert wurde. Die Expression von
FAM26F stieg in CNAR- im Vergleich zu CNAR+ CD8+ T Zellen während
der Kokultivierung mit SIV-infizierten CD4+ T Zellen (viraler
Inhibitionstest) signifikant stärker an. Weitere Untersuchungen
zeigten, dass FAM26F in Blutlymphozyten (CD4+-, CD8+-, B-Zellen)
exprimiert wird. In vitro Stimulations-Studien zeigten, dass die
RNA-Menge von FAM26F 6 Stunden nach der Zugabe von IFN-γ und IFN-α
in PBMCs anstieg. Das Expressionsmuster von FAM26F wurde außerdem
in 2 unabhängigen AIDS-Vakzine-Experimenten vor und nach der
Infektion in insgesamt 42 Rhesusaffen untersucht. In beiden
Experimenten stieg nach der Infektion die Expression von FAM26F in
PBMCs signifikant an und folgte damit einem ähnlichen Muster wie
MX1, IP-10 und Tetherin. Die RNA-Mengen von FAM26F korrelierten
zudem mit denen von MX1, IP-10 und Tetherin. In dem ersten
AIDS-Vakzine-Experiment korrelierten die RNA-Mengen von FAM26F vor
der Infektion invers mit der Virusbeladung im Plasma 2, 12 und 24
Wochen nach der Infektion. In dem zweiten Experiment korrelierte
die Expression von FAM26F 2 Wochen nach der Infektion positiv mit
den viralen RNA Kopien im Plasma 12, 24 und 48 Wochen nach der
Infektion. Die Expression von FAM26F, MX1, IP-10 und Tetherin wurde
außerdem vor und nach der Immunisierung in 2 Gruppen von Tieren
untersucht. Den Tieren wurden als „Boost-Immunisierung“ Vektoren
mit einklonierten SIV-Genen verabreicht, die entweder vom
Gefügelpockenvirus oder vom Adenovirus abgeleitet waren. Impfschutz
wurde in der Gruppe, die mit Adenovirus-Vektoren behandelt wurde,
erzielt und korrelierte mit einer signifikant erhöhten Expression
von IP-10 (24 Std. nach Immunisierung), FAM26F und Tetherin (24 und
48 Std. nach der Boost-Immunisierung). Dies deutet darauf hin, dass
eine verstärkte IFN-γ oder eine bestimmte Balance von Typ I und II
Interferon-Antworten für den Schutzeffekt mitverantwortlich war.
Zusammenfassend zeigen die Ergebnisse, dass FAM26F eine wichtige
Funktion bei der Regulation der angeborenen und adaptiven
Immunantwort spielen könnte. Eine niedrigere Expression von FAM26F
vor der Infektion könnte einen Immunstatus anzeigen, der die frühe
Virusreplikation eingrenzen kann. Eine stärkere Expression von
FAM26F nach der Infektion könnte hingegen auf eine frühe
Dysregulation der Immunantwort deuten, die später mit höherer
Virusbeladung assoziiert ist. Die Expression FAM26F könnte sich
somit als ein früher prognostischer Marker für die SIV/HIV
Infektion erweisen. | de |

dc.description.abstracteng | CD8+ T cell mediated antiviral response
(CNAR) is associated with long-term control of HIV-infection and
attributed to the secretion of an unknown factor called soluble
CD8+ cell antiviral factor (CAF). In order to identify CAF,
microarray data from CD8+ cells displaying high CNAR activity and
CD8+ cells that lack CNAR were analyzed. Out of more than 50
differentially regulated genes, differential expression of 16 genes
was validated by qRT-PCR in CD8 cells from 21 monkeys. FAM26F was
identified as a sole candidate that was significantly
differentially expressed in samples from SIV-infected as well as
non-infected animals. FAM26F expression increased in CNAR- CD8+ T
cells during their co-cultivation with SIV-infected CD4+ T cells in
viral inhibition test. FAM26F was found to be expressed on three
major blood cell populations (CD4+, CD8+ T cells and B cells). In
vitro stimulation studies revealed that FAM26F expression was
greatly induced in PBMCs after 6hrs of IFN-γ stimulation, and to
some extent by IFN-α. Next, the expression pattern of FAM26F before
and after infection was investigated in two independent AIDS
vaccine experiments comprising in total 42 monkeys. In both
experiments, FAM26F expression along with other innate immune
modulators was significantly increased in PBMCs and followed same
in vivo expression pattern after infection as Mx1, IP-10 and
tetherin after SIV-infection. Its expression was also found to be
significantly correlated with Mx1, IP-10 and tetherin. In first
experiment, preinfection RNA levels of FAM26F were inversely
correlated with 2, 12 and 24 wpi viral load while in other
experiment, 2wpi expression of FAM26F was positively correlated
with plasma viral RNA copies at 12, 24 and 48 wpi. Expression of
FAM26F, MX1, IP-10 and tetherin was studied before and after
immunization in two groups of animals that were finally boosted
with fowlpox virus- or adenovirus-derived vector respectively.
Increased level of protectionin adenovirus-derived vector group was
most likely attributed to significantly elevated expression of
IP-10 (24hrs post boosting), FAM26F and tetherin (24 hrs and 48hrs
post boosting) indicating more pronounced IFN-γ responses or a
unique balance between type I and type II responses. In summary,
the results emphasize that FAM26F may be an important regulator of
innate or adaptive immune response. FAM26F expression may be an
early prognostic marker for SIV/HIV infection. Lower expression of
FAM26F before infection may indicate an immune status that is able
to limit early viral replication, whereas a strong increase after
infection may indicate an early immune dysregulation that is later
on associated with higher viral load. Irrespective whether FAM26F
is involved directly in regulation of viral replication or
indirectly via the immune defense; our study has shown that it is
an important molecule that clearly merits further
