dc.description.abstract | Zytotoxische T Zellen können maligne
Tumorzellen durch Freisetzung von Perforin und Granzymen aus
präformierten zytotoxischen Granula lysieren. Im Rahmen der
zytotoxischen Reaktion sind sowohl die Zielzellen als auch die
Killerzellen den Zelltod-Mediatoren ausgesetzt, die Killerzellen
überleben diesen Kontakt allerdings. Ihre Unempfindlichkeit wird
auf eine lokale membrangebundene Expression der sonst lysosomalen
Protease Cathepsin B am Ort der immunologischen Synapse
zurückgeführt, wobei Cathepsin B die Killerzellen wahrscheinlich
durch Spaltung von Perforin vor der Autolyse schützt. Auch
Tumorzellen exprimieren Cathepsin B in membrangebundener Form. Eine
Reduktion ihrer zellmembranassoziierten Cathepsin-B-Aktivität
erhöht dabei die Empfindlichkeit von Tumorzellen gegenüber
Granula-assoziierter Zytotoxizität. Diese Arbeit zeigt, dass die
Überexpression von Cathepsin B allein jedoch noch keinen
Immune-Escape-Mechanismus für Tumorzellen darstellt. Dazu wird die
Cathepsin-B-Expression von RMA-Lymphomzellen durch Transfektion mit
zwei unterschiedlichen Expressionskonstrukten gesteigert und der
Einfluss dieser den Tumorzellen nachempfundenen Überexpression von
Cathepsin B auf die Lyse durch zytotoxische T-Zellen im
Chrom-Freisetzungs-Test analysiert. Zur Untersuchung des
zytoprotektiven Effekts einer Überexpression lysosomaler Cathepsin
B-Formen werden die Zellen mit einem Fusionsprotein aus Cathepsin B
und eGFP unter Kontrolle eines CAG-Promotors transfiziert. Der
Einfluss von membrangebundenem Cathepsin B auf die Lyse durch
zytotoxische T Zellen wird an Zelllinien analysiert, die ein
Fusionsprotein bestehend aus der einkettigen aktiven Form von
Cathepsin B und der Transmembrandomäne von H2K exprimieren. Die
Expression beider Fusionsproteine wird mittels
Durchflusszytometrie, Immunoblot und Laser-Scanning-Mikroskopie
analysiert. Beide Fusionsproteine führen zu einer Steigerung der
intrazellulären Cathepsin B-Expression, eine Assoziation der
Fusionsproteine mit der Zellmembran kann allerdings nicht gezeigt
werden. Der Einfluss der so gesteigerten Cathepsin-B-Expression auf
die Lyse der RMA-Zellen durch zytotoxische T-Zellen wird im
Chrom-Freisetzungs-Test analysiert. Zellen der Klone beider
Konstrukte werden peptidspezifisch durch zytotoxische T-Zellen über
den Granula-Exozytose-Weg lysiert und die Stärke der zytotoxischen
Reaktion am Anteil lysierter Zielzellen gemessen. Hierbei zeigt
sich, dass die Expression der Fusionsproteine durch die RMA-Zellen
keinen Einfluss auf ihre Empfindlichkeit gegenüber zytotoxischen
T-Zellen hatte. Ein Immune-Escape-Mechanismus, der auf der
alleinigen Überexpression von Cathepsin B beruht, kann somit nicht
gezeigt werden. | de |

dc.description.abstracteng | Cytotoxic T cells lyse malignant tumor
cells by releasing perforin and granzymes from preformed cytotoxic
granules. Within the cytotoxic response the target cells and the
killer cells are exposed to cell death mediators. However, the
killer cells usually survive this contact. Their resistance is
attributed to a local membrane-bound expression of the otherwise
lysosomal protease cathepsin B at the location of the immunological
synapse. Cathepsin B protects the killer cells by cleavage of the
cell death mediator perforin. Tumor cells express cathepsin B in a
membrane-bound form as well. A reduction in their cell membrane
associated cathepsin B activity increases their susceptibility to
granule-associated cytotoxicity. This study shows that
overexpression of cathepsin B alone is not an immune escape
mechanism for tumor cells. The cathepsin B expression of RMA
lymphoma cells was enhanced by transfection with two different
expression constructs and the influence of this tumor cell like
overexpression of cathepsin B on the lysis by cytotoxic T-cells was
analyzed in chromium release assay. To examine a cytoprotective
effect of an overexpression of lysosomal forms of cathepsin B, the
RMA lymphoma cells are transfected with a fusion protein consisting
of cathepsin B and eGFP under control of a CAG promoter. The
influence of membrane-bound cathepsin B on tumor cell lysis by
cytotoxic T cells is analyzed in RMA cell lines expressing a fusion
protein consisting of a single-chain active form of cathepsin B and
the transmembrane domain of H2K. The expression of the two fusion
proteins was analyzed by flow cytometry, immunoblotting and
laser-scanning microscopy. Both of the fusion proteins lead to an
increase in intracellular cathepsin B expression. However, an
association of the fusion proteins with the cell membrane cannot be
shown. The influence of the increased cathepsin B expression on the
lysis of RMA cells by cytotoxic T cells is analyzed in the chromium
release assay. Cells of clones of both constructs are lysed by
peptide-specific cytotoxic T-cells via the granule exocytosis
pathway and the strength of the cytotoxic response is measured as
proportion of lysed target cells. It is shown that the expression
of the fusion proteins does not affect the sensitivity of RMA
lymphoma cells to cytotoxic T cells. An immune escape mechanism
based solely on the overexpression of cathepsin B can therefore not
