mittels semi-quantitativen real-time-PCR in kolorektalen
Methodik legen. | de |

dc.description.abstracteng | Colorectal cancer originates from normal
mucosa cells as a result of genetic alterations that cause changes
in their differentiation status and in the control of cell growth.
Understanding the molecular mechanisms of colorectal cancer
initiation and progression is therefore exceedingly important for
the development of novel therapeutic strategies. The aim of this
study was to identify genes that may impact the growth and
proliferation of colorectal cancer cells. Based on prior gene
expression analyses of colorectal cancers and matched normal mucosa
samples, a set of six genes that were expressed at very low levels
in colorectal cancers compared to the mucosa samples was selected
for further experiments. First, the expression of these genes was
assessed in colorectal cancer cell lines using semi-quantitative
real-time PCR. Two of these genes, NR3C1 and PDK4, were then
overexpressed in the colon cancer cell line SW480 using an
inducible vector system. After single cell cloning, Western blot
analysis was performed in order to identify those clones with the
highest protein expression for functional analysis. Follow-up
experiments revealed that the cellular viability of these clones
was not affected, but the ability to form colonies was reduced.
These findings suggest a potential functional relevance of PDK4 and
NR3C1 for colorectal cancer progression and may form the basis for
further investigations based on the established methods. | de |
