dc.description.abstract | Antikörperabhängige zellvermittelte
Zytotoxizität (ADCC) ist einer der Hauptwirk-mechanismen
monoklonaler anti-CD20-Antikörper bei der Elimination maligner
B-Zell-Lymphome. Trotz moderner Immunchemotherapien gibt es
Therapieversager. Neue anti-CD20-Antikörper mit Modifikationen am
Fc- und Fab-Teil könnten die Heilungsrate verbessern und die
Rezidivrate verringern. Zu ihnen zählen GA101 und Ofatumumab. In
einem Lymphom-Modell mit NK-Zellen als Effektorzellen und
verschiedenen B-Lymphomzellen als Zielzellen wurde die Effizienz
der antikörperabhängigen zellvermittelten Zytotoxizität zwischen
Rituximab, Ofatumumab und GA101 verglichen. Dies ist der erste
direkte Vergleich zwischen den drei Antikörpern Rituximab,
Ofatumumab und GA101. Dieser Vergleich zeigt auf, dass GA101
hinsichtlich der ADCC Rituximab überlegen ist. Ofatumumab ist
Rituximab hinsichtlich der ADCC ebenbürtig. Der neu entwickelte
Dritt-generations-Antikörper GA101 besitzt eine höhere Effizienz in
der Vermittlung der NK-Zell- abhängigen Zielzelllyse. Der Grund für
die verbesserte Zielzelllyse ist die durch GA101 gesteigerte
NK-Zell-Aktivität. GA101 ist nicht nur effizienter im Bereich
niedriger Antikörper-konzentrationen, sondern zeigt auch bei
einigen Lymphomzelllinien eine größere maximale Zielzelllyse und
NK-Zell-Aktivität. Sowohl die NK-Zell-Aktivität als auch die
zytolytische Effizienz konnten bei allen drei Antikörpern
konzentrationsabhängig gesteigert werden. Die mittlere effektive
Konzentration, die zu einer halbmaximalen NK-Zell-Aktivität (EC50)
führte, lag bei GA101 in einem Bereich, der etwa eine Zehnerpotenz
niedriger war als bei Rituximab und Ofatumumab. Die Versuche zur
direkten Zytotoxizität monoklonaler Antikörper in dieser Arbeit
zeigen die geringe Bedeutung einer Antikörperwirkung ohne
Effektorzelle auf. Durch die Verbesserungen der ADCC stellt GA101
bei Hochrisikopatienten, bei Patienten mit Krankheitsprogress und
im Rezidiv eine vielversprechende neue Therapieoption dar. In
weiterführenden klinischen Studien gilt es, dieses Ergebnis in vivo
zu prüfen und Patientengruppen zu definieren, die potentiell am
stärksten von einer Therapie mit GA101 profitieren. | de |

dc.description.abstracteng | Antibody-dependent cell-mediated
cytotoxicity (ADCC) is one of the major effector mechanisms of
anti-CD20 antibodies in eliminating B-Cell lymphomas. Despite
modern immunochemotherapies relapses occur. Novel anti-CD20
antibodies with Fab and Fc fragment modifications are expected to
overcome resistances and to increase the cure rate. Ofatumumab and
GA101 are modified anti-CD20 antibodies. This work contains an in
vitro model of human natural killer (NK) cell-mediated ADCC against
human B lymphoma cell lines comparing Rituximab, Ofatumumab and
GA101. This is the first direct comparison of Rituximab, Ofatumumab
and GA101. This comparison shows that the novel antibody GA101 is
more effective in mediating ADCC than Ofatumumab and Rituximab.
Ofatumumab and Rituximab are equally potent in mediating ADCC. The
novel third generation antibody GA101 has a higher potency in NK
cell-mediated lysis. The reason for the higher lysis of target
cells is the enhanced NK cell activity. GA101 is not only more
potent in the range of low concentration, but shows also a higher
maximum lysis and NK cell activity for some cell lines. NK cell
stimulation and the resulting cytolytic efficacy measured by target
cell lysis as well as the NK cell activity were anti-CD20 antibody
concentration dependent for Rituximab, Ofatumumab and GA-101. The
half maximal effective concentration (EC50) for the NK cell
activity was 10 times lower for GA101 in comparison to Ofatumumab
and Rituximab. The data of this work concerning direct cytotoxicity
(PCD) show the unimportance of this mechanism in the elimination of
B-cell lymphoma cells in vitro. The enhancement of ADCC by GA101
could be beneficial for high risk patients, patients with progress
or relapse. Further studies will be needed to define the group of
patients who will benefit the most. | de |
