dc.description.abstract | Spinosyn A und D sind die beiden Hauptmetabolite der
Streptomyces-Spezies Saccharopolyspora spinosa. Sie weisen eine
hohe Aktivität sowie Selektivität gegenüber einer Vielzahl von
Schadinsekten auf. Aufgrund ihres hervorragenden ökologischen
Profils werden die Verbindungen nicht nur im Pflanzenschutz sondern
auch in der Veterinär- und Humanmedizin zum Schutz von Menschen,
Nutz- und Haustieren vor Parasiten verwendet. Die aktive Verbindung
Spinosad wird durch Fermentation gewonnen und ist eine Mischung von
Spinosyn A und D.
Durch das Auftreten erster Resistenzen bei vereinzelten
Schädlingsklassen ist im Rahmen eines verantwortungsvollen
Resistenzmanagements die Bereitstellung neuartiger Spinosynanaloga
dringend erforderlich. Hierbei könnte die organische Synthese eine
wichtige Rolle spielen. Die literaturbekannten Totalsynthesen sind
jedoch für die industrielle Anwendung aufgrund von geringer
Effizienz und Wirtschaftlichkeit nicht geeignet.
Ziel dieser Arbeit war die Entwicklung neuer Bausteine für eine
effiziente, konvergente Syntheseroute zum Aufbau von
Spinosynanaloga mit einem modifizierten tricyclischen Grundgerüst.
Darüber hinaus sollten erste Untersuchungen zur Kupplung dieser
Bausteine durchgeführt werden. | de |

dc.description.abstracteng | The Spinosyns A and D are the main
metabolites of the Streptomyces species Saccharopolyspora spinosa.
They show a high activity as well as a high selectivity against a
huge number of insects that cause extensive damage to crops and
other plants. Because of their excellent environmental profile
these compounds are not only used in crop protection but also in
veterinary and human medicine to protect humans, livestock and
companion animals from external parasites. The active compound
Spinosad is a fermentatorically produced mixture of Spinosyn A and
D. First signs of resistances of some target insects make the
production of novel Spinosyn analogues inevitable. Here the organic
synthesis could play an important role. Because of their low
efficiency and profitability the reported total syntheses are,
however, not suitable for an industrial application.
The main goal of this thesis was the development of new building
blocks for an efficient and convergent route for the synthesis of
Spinosyn analogues bearing a modified tricyclic framework.
Furthermore first investigations towards the coupling of these
building blocks were conducted. | de |
