dc.description.abstract | Die vorliegende Dissertation befasst sich
mit der Darstellung lipidierter und nicht-lipidierter
3-L-Hydroxyleucin-Derivate, sowie der Darstellung verschiedener
Derivate und Analoga der Lipidseitenkette von Muraymycin A1. Im
Rahmen der 3-L-Hydroxyleucin-Synthese werden verschiedene Zugänge
zu N- und C-terminal funktionalisierbaren Bausteinen für die
Totalsynthese des Muraymycin-Rückgrads gezeigt. Die Lipidierung der
Aminosäure wird in Modellstudien mittels Veresterung sowie
Olefinmetathese durchgeführt. In der Dissertation werden des
Weiteren verschiedene synthetische Zugänge zur Darstellung der
N-Alkyl-N-hydroxyguanidin-Funktionalität dargestellt und
verglichen. Als Nachweis der breiten Anwendbarkeit der
schutzgruppenfreien Synthesestrategie wird die Darstellung der
nicht-proteinogenen Aminosäure N5-Hydroxyarginin sowie weiterer
N-Alkyl-N-hydroxyguanidine gezeigt. Die Synthese eines
fluoreszenzmarkierten Derivats der Muraymycin A1-Lipidseitenkette
liefert im Rahmen fluoreszenzmikroskopischer Untersuchungen an
unilamellaren Riesenvesikeln erste Hinweise auf die biologische
Rolle dieser Muraymycin-Substruktur. Abschließend werden erste
Arbeiten zur Synthese von Analoga der Lipidseitenkette mit
bestimmter Konformation gezeigt. | de |

dc.description.abstracteng | The present thesis deals with the
synthesis of lipidated and non-lipidated 3-L-hydroxyleucin
derivatives as well as the synthesis of different derivatives and
analogues of the lipid side chain of muraymycin A1. Within the
3-L-hydroxyleucin synthesis different approaches for N- and
C-terminal derivatisable building blocks for the total synthesis of
the muraymycin backbone are shown. The lipidation of the amino acid
is achieved through esterification and olefin metathesis.
Furthermore different synthetic approaches for the synthesis of the
N-alkyl-N-hydroxyguanidine functionality are presented and
compared. As proof of principle for the protection group free
approach the syntheses of the non-proteinogenic amino acid
N5-hydroxyarginine as well as further N-alkyl-N-hydroxyguanidines
are shown. The synthesis of a fluorescence-labeled muraymycin A1
lipid side chain analogue and subsequent fluorescence microscopy
using giant unilamellar vesicles yield first insights into the
biological role of this muraymycin substructure. As a last point
first attempts in the synthesis of lipid side chain analogues with
a predetermined conformation are shown. | de |
