dc.description.abstract | Die Expression und Aktivität der
Ca2+/Calmodulin-abhängigen Proteinkinase IIδ (CaMK IIδ) ist in
Kardiomyozyten insuffizienter Herzen erhöht und geht mit einer
Krankheitsprogression einher. Ebenso führen Reaktive
Sauerstoffspezies, die während einer Ischämie oder
Reperfusionsphase vermehrt gebildet werden, zu einer Natrium- und
Kalziumüberladung und letztendlich zu kontraktiler Dysfunktion und
zum Zelltod. Anhand dieser Arbeit kann gezeigt werden, dass die
sauerstoffradikalinduzierte Toxizität über die
Ca2+/Calmodulin-abhängige Proteinkinase IIδ vermittelt wird bzw.
die aktivierte Ca2+/Calmodulin-abhängige Proteinkinase IIδ für die
Entstehung von oxdativen Zellschäden essentiell ist.
Kardiomyozyten, in denen das Gen für die CaMKIIδ ausgeschaltet ist
(CaMKIIδ-/-), weisen eine deutlich verzögerte ROS-induzierte
Zunahme der zytosolischen Natrium- und Kalziumkonzentration auf und
präsentieren eine signifikant geringere arrhythmogene Aktivität
unter Radikalexposition. In Wildtyp-Kardiomyozyten wird die
Kalziumüberladung hauptsächlich über den Umkehrtransportmodus des
sarkolemmalen Natrium-Kalzium-Austauschers vermittelt, der bei
hohen zytosolischen Natriumkonzentrationen Kalzium in die Zelle
transportiert. Dies kann durch den ausbleibenden Kalziumanstieg in
mit KB-R7943 behandelten Myozyten bestätigt werden. Letztendlich
schützt der homozygote Knock-out der CaMKIIδ durch eine
ausbleibende ROS-induzierte Natriumakkumulation vor einer Umkehr
der Transportrichtung des Natrium-Kalzium-Austauschers und damit
konsekutiv vor einer Kalziumüberladung der Zelle. Dies geht mit
einer verminderten Hyperkontrakturentwicklung einher und führt
damit zu signifikanten Überlebensvorteilen durch das Ausschalten
der CaMKIIδ. In Anlehnung an die gewonnenen Erkenntnisse könnte
eine gezielte pharmakologische Inhibition der CaMKIIδ daher
möglicherweise eine völlig neuartige Therapiestrategie in der
Behandlung der Herzinsuffizienz darstellen. | de |

increased in heart failure and correlates with progression of
disease. Also, reactive oxygen species (ROS), which are
increasingly produced during the ischaemic or reperfusion phase,
lead to an overload with sodium and calcium and result in
contractile dysfunction and apoptosis. This work illustrates that
ROS-induced cellular toxicity is mediated by the
Ca2+/Calmodulin-dependent protein kinase IIδ and that activated
Ca2+/Calmodulin-dependent protein kinase IIδ is essential for the
development of oxidative cell damage.
CaMKIIδ-knock-out-Cardiomyocytes (CaMKIIδ-/-) show a delayed
ROS-induced increase in cellular sodium and calcium concentration
and represent a significantly lower arrhythmogenic activity under
ROS exposition. In wild type cardiomyocytes the calcium overload is
mainly mediated by the reverse mode of the sarcolemmal
sodium-calcium-exchanger which transports calcium in the cell
during high cellular sodium concentrations. This can be confirmed
by the lack of sodium increase in KB-R7943 treated myocytes.
Ultimately, the homozygous knock-out of the CaMKIIδ protects from a
reversal of transport direction of the sodium-calcium-exchanger by
stopping the ROS-induced sodium accumulation. Consecutively this
protects from a calcium overload of the cell, goes along with a
decreased development of hypercontractures and a significant
survival advantage by disabling the CaMKIIδ. Following these
discoveries, targeted pharmacological inhibition of the CaMKIIδ
might illustrate entirely new therapeutic strategies in the
treatment of heart failure. | de |
