dc.description.abstractImmer älter werdende Menschen, insbesondere Frauen in der Menopause, können durch den zunehmenden Mangel an E2, T und IGF-1 von bis zu 1 % pro Lebensjahr (Bauer et al. 2008) an einer Sarkopenie erkranken. Epidemiologische Studien belegen, dass etwa 5-13 % aller 60- bis 70-Jährigen und bis zu 50% der über 80-Jährigen von Sarkopenie betroffen sind und diese Erkrankung somit eine zunehmend soziologische und ökonomische Rolle spielt. In dieser Studie wurde untersucht, ob bei ovx-Ratten die Gabe von Ecd einen positiven Effekt auf die Muskulatur hat und somit eine Therapiealternative neben Testosteron und Estrogenapplikation für postmenopausale Frauen zur Verhinderung einer altersabhängigen Sarkopenie darstellt. Des Weiteren sollte geklärt werden, ob das eventuell anabol auf die Skelettmuskulatur wirkende Ecd eine Herzmuskelfaserzunahme bewirkt und die Lipidproteine im Blut verändert, um eventuell eine Vorhersage in Bezug auf kardiovaskuläre Krankheiten treffen zu können. Hierzu wurden zwei voneinander zeitlich getrennten Versuchen durchgeführt. Im ersten Versuch ging es um eine subkutane (s.c.) Gabe der Testsubstanz in verschiedenen Konzentrationen über einen Zeitraum von vier Wochen. Im zweiten Versuch wurde die Testsubstanz oral in Pellettform in verschiedenen Konzentrationen für drei Monate lang verabreicht. In beiden Versuchen handelte es sich bei den Versuchstieren um insgesamt 142 weibliche Sprague-Dawley-Ratten 61 Ratten für den ersten und 81 Ratten für den zweiten Versuch. Am Ende der Versuchsreihen wurde die Tibia der Tiere mittels einer quantitative Computertomographie (qCT) gemessen, der M. gastrocnemius und das Herz histologisch ausgewertet. Zusätzlich wurden die Serumparameter von IGF-1 sowie Lipoproteine der Tiere bestimmt. Dadurch zeigte die Studie, dass die anabole Wirkung der Substanz Ecd auf die Skelettmuskulatur auch bei ovarektomierten Ratten abhängig von der Applikationsform sichtbar ist, ohne dabei die Herzmuskulatur negativ zu beeinflussen, indem es nicht wie anabole Steroide eine hypertrophe Kardiomyopathie verursacht hatte. Die Substanz Ecd beeinflusst den Lipoproteinstoffwechsel, indem es HDL erhöht und minimal LDL senkt. Daher könnte die Substanz Ecd einen protektiven Effekt auf die Entstehung von KHK haben.de

Special women in the portmenopausal period with an increasing deficit of E2, T and IGF-1 from 1% each year of one¡¯s life could get an sarcopenia. Many Studies presented that about 5-13% of all 60 till 70 years old people and 50% of all 80 years old people are affected by sarcopenia and that means an high social and economic Problem.  This study should show, if Ecd is donated to ovx-Rats, that they have an positive effect on the skeletal muscle as a kind of alternative method next to testosterone and estrogen at postmenopausal women's without having the side effects like kardiomyopatia or a negative effect at the lipoprotein-serum.  For evaluation we took 2 different tests. One test with subcutane injection (s.c.) for four weeks long and one test where the rats became the test-substance oral as a Pelletform in different concentrations for 3 months. In each test we took ovx Sprague- Dawley- Rats. 61 rats at the first test and 81 rats at the second test. After the finishing we analyzed the muscle at the tibia with a qCT, took bloodsamples of IGF-1, HDL and LDL and examinated histological the M. gastrocnemius and the heart. Thereby this study showed, that the anabol working substance Ecd has an effect at the skeletal muscle at ovx rats depending in different application forms without having the negative side effects at the heart like anabolic steroids. The substance Ecd affected an increasing of HDL and a small decreasing of LDL at the blood serum.  That means that the anabolic working substance Ecd could defend postmenopausal women¡¯s for sarcopenia and protect them for cardiavascular diseases.de
