dc.description.abstract | Prostaglandin (PG) E2 und IL-4 sind an der
Reifung von DC und an der Modulation der DC-Migration beteiligt. In
der vorliegenden Studie konnte gezeigt werden, dass IL-4 und IL-13
die Wanderung von murinen DC und Makrophagen gegen verschiedene
inflammatorische Mediatoren beeinträchtigen. Dabei wurde die
Migration in einem syngenen in vivo Mausmigrationsmodell gegen die
Chemokine MIP-1alpha (CCL3), MIP-3alpha (CCL20), MCP-1 (CCL2) und
MDC (CCL22), gegen das N-formyl Peptid fMLP und das antimikrobielle
Peptid LL-37 und gegen die Anaphylatoxinen C5a und C3a durch IL-4
und IL-13 kompromittiert. Zusätzlich wurde eine STAT6- und
PI3K-abhängige Reduktion der Oberflächenexpression von C5aR, C3aR,
CCR2 und CCR5 nachgewiesen. IL-4 beeinträchtigte ebenfalls die
Chemotaxis von humanen Monozyten und von aus Monozyten generierten
DC (MoDC) gegen MIP-1alpha und MCP-1 in einem
SCID-Mausmigrationsmodell und regulierte die Expression von CCR1,
CCR2, CCR5 auf Monozyten und CCR2, aber nicht CCR1 und CCR5 auf
MoDC herunter. Der negative Einfluss von IL-4 auf die Migration von
Monozytenabkömmlingen gegen MIP-1alpha wurde durch PGE2 verstärkt.
Dabei verringerte PGE2 aber zusätzlich die Oberflächenexpression
von CCR1 und CCR5. PGE2 wirkte der IL-4-induzierten
Herunterregulierung der Expression und Funktion des C5a-Rezeptors
(C5aR) auf Monozyten und MoDC entgegen. Im Gegensatz dazu
verringerte in einem SCID-Mausmodell PGE2 in Kombination mit
TNF-alpha die C5aR-Expression auf Monozyten während ihrer schnellen
Differenzierung in reife CCR7-exprimierende DC. Zusammenfassend
zeigen die Daten der vorliegenden Studie, dass das TH2-Cytokin IL-4
allgemein die Kapazität der DC und Monozyten/Makrophagen gegen
Chemokine, Anaphylatoxine, Formylpeptide und antimikrobielle
Peptide zu wandern, kompromittiert und PGE2 hingegen eine
komplexere Rolle in der Regulation der DC-Chemotaxis zu spielen
scheint und in unterschiedlicher Weise die Rezeptorexpression in
vitro und in vivo beeinflusst. | de |

dc.description.abstracteng | Prostaglandin (PG) E2 and IL-4 have been
implicated in maturation of DC and in the modulation of DC
migration. In the present study, IL-4 as well as IL-13 were found
to impair migration of murine DC and macrophages in response to
various inflammatory mediators such as the chemokines MIP-1alpha
(CCL3), MIP-3alpha (CCL20), MCP-1 (CCL2), MDC (CCL22), the N-formyl
peptide fMLP, the antimicrobial peptide LL-37, and the
anaphylatoxins C5a and C3a in an syngeneic in vivo mouse migration
model. Concomitantly, a STAT6- and PI3K-dependent reduction of
C5aR, C3aR, CCR2 and CCR5 surface expression was detected. IL-4
also impaired the chemotaxis of human monocytes and
monocyte-derived DC toward MIP-1alpha and MCP-1 in an SCID mouse
migration model and downregulated expression of CCR1, CCR2, CCR5 on
monocytes and CCR2 but not CCR1 and CCR5 on monocyte-derived DC.
The negative impact of IL-4 on the migration of monocyte-derived
cells toward MIP-1alpha was enhanced by PGE2 which additionally
decreased the surface expression of CCR1 and CCR5. However, PGE2
counteracted the IL-4-induced downregulation of C5a receptor (C5aR)
expression and function in monocytes and monocyte-derived DC. In
contrast, PGE2 abrogated C5aR expression in monocytes while
inducing their rapid differentiation into mature CCR7-expressing DC
in a SCID mouse model in combination with TNF-alpha. In summary,
the data of this study indicate that the Th2 cytokine IL-4
generally compromised the capacity of DC and monocytes/macrophages
to migrate toward chemokines, anaphylatoxins, formyl as well as
antimicrobial peptides whereas a more complex role in the
regulation of DC chemotaxis is suggested for PGE2 which
differentially affected chemotactic receptor expression in vitro
and in vivo. | de |
