dc.description.abstract | Die embryonale Entwicklung der vom
primitiven Darmrohr abgeleiteten Organe folgt in Vertebraten
konservierten molekularen Mechanismen. Im Laufe der
Embryonalentwickung differenzieren sich von anterior nach posterior
das ösophagial-trachiale System, der Magen, das Duodenum mit den
anliegenden Organen Leber, Pankreas und Gallenblase, sowie der
Dünn- und der Dickdarm. Der Wnt Signaltransduktion scheint eine
entscheidende Rolle in diesen Prozessen zuzukommen. Im Rahmen
dieser Arbeit wurde die Rolle des sekretierten Wnt Antagonisten
XsFRP5 auf die Entwicklung entodermaler Organe in Xenopus Embryonen
untersucht. Bei XsFRP5 handelt es sich um einen Wnt Antagonisten
der „secreted Frizzled-related Proteins“ (sFRPs). Es konnte gezeigt
werden, dass XsFRP5 sowohl kanonische als auch nicht-kanonische Wnt
Liganden binden und inhibieren kann. Bestimmte Xwnt Proteine wurden
von XsFRP5 zwar effizient gebunden, jedoch kaum in ihrer Funktion
beeinträchtigt. Der „Knock-down“ von XsFRP5 resultierte in einer
Reduktion der ventralen Pankreasanlage sowie einer Expansion des
Magens. Die Überexpression von XsFRP5 in VegT-programmierten
animalen Kappenexplantaten induzierte die robuste Induktion
pankreatischer Markergene, wobei dieser Effekt nicht nur durch eine
Inhibition der Wnt Signaltransduktion, sondern auch über eine
hiervon abhängige Induktion der Retinsäure-Signalwirkung bedingt
ist. Der Vergleich von XsFRP5 mit dem strukturell verwandten Wnt
Antagonisten XsFRP2 ergab, dass es sich bei der Induktion
pankreatischer Markergene um einen XsFRP5-spezifischen Effekt
handelt. Es scheint wahrscheinlich, dass XsFRP5 die Auftrennung der
hepato-pankreatischen Vorläuferzellpopulation in Leber und
ventrales Pankreas beeinflusst. Gleiches gilt für die Grenze
zwischen Magen und Duodenum, die sich nach Verlust von XsFRP5 zu
verschieben scheint. Die im Rahmen dieser Arbeit durchgeführten
Interaktionsstudien, gemeinsam mit der Analyse der
Expressionsmuster, definieren Xwnt2b und Xwnt5a als wahrscheinliche
in vivo Zielproteine von XsFRP5. | de |

dc.description.abstracteng | In vertebrates the embryonic development
of all endodermal organs follows conserved molecular mechanisms.
Wnt signaling is supposed to play a key role during these
processes. The gastrointestinal tract comprises the oesophagus, the
trachea, the stomach, the duodenum and its associated organs liver,
pancreas and gall bladder as well as the small intestine and the
colon. In this study the role of the secreted Wnt antagonist XsFRP5
during the development of the gastrointestinal tract was analysed
in Xenopus embryos. XsFRP5 belongs to the family of „secreted
Frizzled related proteins“ (sFRPs). XsFRP5 is a mutlivalent Wnt
antagonist, which is able to bind and inhibit canonical as well as
non-canonical Wnt proteins. After depletion of XsFRP5 protein by
morpholino injection the ventral pancreatic anlage was reduced,
whereas the stomach seemed to be enlarged. Overexpression of VegT
and XsFRP5 in animal cap explants resulted in a strong and
dose-dependent induction of pancreatic marker genes, an effect that
also depends on retinoic acid signaling. The structurally related
XsFRP2 is not able to mimick the gain-of-function effects in the
animal cap system, arguing for a specific function of XsFRP5 in
pancreas development. It seems likely that XsFRP5 regulates the
border delineation between liver and ventral pancreatic anlage as
well as between stomach and duodenum. Combining data obtained from
interaction studies and expression analysis led us to conclude that
Xwnt2b and Xwnt5a are the most likely in vivo targets of
XsFRP5. | de |
