dc.description.abstractUnser Labor konnte kürzlich zeigen, dass ein einstündiger Immobilisationssstress in einer verbesserten Lernfähigkeit resultiert, wenn die Mäuse 3 h später mittels Furchtkonditionierung trainiert wurden. Weiterhin konnte gezeigt werden, dass das laterale Septum und der Hippokampus an der Regulation der stress-verbesserten Furchtkonditionierung beteiligt sind. Es wurde daher vermutet, dass eine veränderte Genexpression im lateralen Septum und im Hippokampus für stress-verbessertes Lernen verantwortlich sein könnte. Mittels Subtraktiver Hybridisierung konnte zu dem Zeitpunkt 30 min nach dem Ende der Immobilisierung eine Aufregulation für das Gen der Cyclin abhängige Kinase (Cdk) 5 beobachtet werden. Cdk5 ist eine Ser/Thr Kinase und wurde ursprünglich aufgrund seiner Homologie zu der Zellzyklus Kinase Cdk2 identifiziert. Allerdings ist Cdk5 nicht an der Regulation des Zellzyklus beteiligt, sondern bisher vor allem in Zusammenhang mit embryonalen Entwicklungsprozessen, wie der Axon-Migration beschrieben worden. Um die physiologische Funktion der Cdk5 im adulten Gehirn zu untersuchen, wurden die zelluläre Lokalisation und die Aktivität der Cdk5 in Abhängigkeit von Immobilisatiosstress und Furchtkonditionierung analysiert. In Antwort auf Immobilisatiosstress und Furchtkonditionierung konnte eine transiente Aufregulation des Cdk5 Protein in cholinergen Neuronen des septo-hippokampalen Systems beobachtet werden. Außerdem konnte gezeigt werden, dass Cdk5 im septo-hippokampalen System mit seinem Aktivatorprotein p35 assoziiert ist. In Einklang damit resultierte der transient erhöhte Cdk5 Proteingehalt in entsprechend höherer Cdk5 Kinaseaktivität. Durch spezifische Injektion von Cdk5 Inhibitoren in Hirnregionen konnte gezeigt werden, dass eine septo-hippokampale Cdk5 Aktivität die Gedächtniskonsolidierung, deren stress-vermittelte Verbesserung und derer Resistenz gegenüber Extinktion reguliert. Somit konnte gezeigt werden, dass Cdk5 ein zentraler Regulator kognitiver Hirnfunktionen ist.de

dc.description.abstractengOur laboratory has recently demonstrated that 1 h of stressful immobilization of Balb/c mice result in an enhanced learning if animals are trained 3 later in the fear conditioning paradigm. Furthermore it was shown that the lateral septum and the hippocampus are involved in this stress-mediated enhancement of fear conditioning. Based on this data it was investigated whether differential gene-expression in the hippocampus and intermediate lateral septum is responsible for stress-enhanced learning. By subtractive hybridization the gene coding for cyclin dependent kinase (Cdk) 5 was found to be up-regulated in the lateral septum of Balb/c mice 30 min after the end of 1 h immobilization. Cdk5 is a Ser/Thr Kinase and was originally identified by its homology to the cell cycle regulating kinase Cdk2. However, Cdk5 is not involved in the cell cycle regulation, but mainly required for developmental processes, such as axon migration. To investigate the physiological role of Cdk5 in the adult brain the cellular localization and activity of Cdk5 was analyzed in response to stress and fear conditioning. The Cdk5 protein was found to be transiently up-regulated in cholinergic neurons in the septo-hippocampal system of Balb/c mice in response to stressful immobilization and fear conditioning. Furthermore it was shown that Cdk5 associates with its activator subunit p35 in the septo-hippocampal system. Consequently, the up-regulation of Cdk5 protein was accompanied by increased Cdk5 kinase activity. By region-specific microinjection of Cdk5 inhibitors it was demonstrated that septo-hippocampal Cdk5 activity was required for memory consolidation, its enhancement by stress and its resistance to extinction. On this basis, the presented work showed that Cdk5 is a central regulator of cognitive brain functions.de
