Osteoporosis as a systemic disease has increased in importance in recent years. One reason is the demographic development. In Germany, about 8 million people are affected (especially women after menopause). The postmenopausal osteoporosis induced by estrogen deficiency was treated with hormone replacement therapy. But rethinking is necessary after side effects become known. The aim of this study was to analyze a potential osteoprotective effect of Tinospora cordifolia (0,26 and 2,6 mg/day), 20-hydroxyecdysone (59,16 mg/day) and STX (0,25 mg/day) compared to 17β-estradiol (0,13 mg/day) in 60 ovariectomized Sprague-Dawley rats. The cortical bone, epiphyseal plate and articular cartilage thickness of the metaphysis of the tibia was measured. The parts of fat, trabeculae and hematopoietic lymphoid tissue were determined. The strut analysis was examined. The measurements of the cortical bone showed no significant differences between all groups. The test substances TC, Ecd and STX showed a significantly higher epiphyseal plate than the control group. Animals under estradiol gift did not share the results. In all animals the cartilage thickness was significantly increased in comparison to the control group, except those under STX gift. The fat content was significantly lower in all test groups compared to the control group. The trabecular bone percentage was significantly higher in all test groups compared to the control group. The proportion of hematopoietic lymphoid tissue was similar in all groups. These results demonstrate the osteoprotective effect of the test substances. Also the strut analysis could confirm this. In summary it was shown that the test substances Tinospora cordifolia, 20-hydroxyecdysone and STX have a similar bone protective effect like 17β-estradiol.
