dc.description.abstracteng | The objective of this project was to perform an expression analysis of the gap junction structural proteins 26, 43 and 45 in oral squamous cell carcinoma of the DMBA-induced cheek pouch carcinoma of the hamster. Immunohistochemical staining of paraffin tissue sections with subsequent quantitative evaluation of the tissue areas examined was used to identify connexins 26, 43 and 45. In addition, the localisation of the connexins on the nucleus, plasma and membrane was determined semi-quantitatively. In this respect, a classification was made according to the intensity of the coloration using microscopic examination. Diagnosis of the malignant transformation was ensured by additional hematoxylin and eosin staining of specimens from the same sequence used for the connexin immunohistochemical staining.
During the course of malignant transformation, a significant under-expression was determined for the three connexin subtypes, respectively. A significant difference in the distribution in the cytoplasm could be determined for connexin 26 and connexin 45. However, no differential connexin localisation on the nucleus and membrane could be diagnosed as part of oral carcinogenesis.
In summary, it can be concluded from this work that oral carcinogenesis is associated with regulation of connexins 26, 43 and 45 at the protein level. | de |
