dc.description.abstracteng20-hydroxyecdysone (Ecd) has numerous favorable effects on a variety of organs, including the skin, where it improves wound healing, and serum lipids. It is devoid of estrogenic and androgenic effects. Therefore, application of 20-hydroxyecdysone might be a new approach to improve skin and decline symptoms of the metabolic syndrome in elderly sex hormone deficient patients, and this was investigated in gonadectomized male and female rats. They were orally treated with Ecd (11, 22, or 67 mg/animal/day for the male and 4 or 11 mg/animal/day for the female) for 12 weeks, and skin samples were evaluated histologically to quantify two dermal layers, the subcutaneous fat and muscle layers. Compared to non-treated rats, epidermal thickness was higher in the high-dose Ecd-treated male rats and lower in the low-dose Ecd-treated rats. Dermal thickness was lower in the male treated rats and higher in the female treated rats. The subcutaneous fat layer was thickest in the gonadectomized rats and thinner in all Ecd-treated animals. The muscle layer was thinner in the high-dose Ecd-treated male rats and unaffected in the female group. The number of proliferating cell nuclear antigen antibody positive cells was significantly higher in all other groups. Ecd application showed no side effects on the serum lipids. In conclusion Ecd has an decreasing effect on subcutaneous fat in male and female and may be able to prevent the metabolic syndrome. The stimulatory effects of Ecd on epidermal cell pofileration results in higher epidermal cell conversion.de
