dc.description.abstractgerZIEL Das Ziel dieser in-vitro-Untersuchung war der Vergleich der Biokompatibilität und der antimikrobiellen Effekte eines mit einem Kupferadditiv modifizierten Zinkoxidphosphatzementes im Vergleich zu einem konventionellen Zinkoxidphosphatzement. MATERIAL/METHODE Zwei Materialien wurde in dieser Studie verglichen (Hoffmann´s Kupfer Cement und Hoffmann´s Cement, Hoffmann Dental Manufaktur, Deutschland). Es kamen solide Probekörper zum Einsatz. Die Versuche starteten mit der Messung des arithmetischen Mittels der Rauheit, der freien Oberflächenenergie und der Oberflächenlokalisation von Kupfer. In den mikrobiellen Untersuchungen folgte die Inkubation mit Streptococcus sanguinis und Candida albicans sowie deren Auswertung mittels Fluoreszenz-Assays. Die Untersuchungen auf Zytotoxizität wurden mit einem CCK8-Assay auf GF1 humanen Gingivafibroblasten und L929 Mausfibroblasten durchgeführt. Die Tests schlossen mit lichtmikroskopischen und rasterelektronischen Aufnahmen der besiedelten Probekörper. ERGEBNISSE Es gab keine relevanten signifikanten Unterschiede der freien Oberflächenenergie, der Rauheit, der Zytotoxizität oder der antimikrobiellen Wirkung. Beide Zemente zeigten jedoch eine hohe zytotoxische Wirkung per se. Die oberflächliche Kupferlokalisation war gering. SCHLUSSFOLGERUNG Das Kupferadditiv zeigte keine signifikanten antimikrobiellen Wirkungen, beide Zemente zeigten jedoch eine hohe Zytotoxizität. Alle weiteren getesteten Parameter zeigen ebenfalls keine relevanten signifikanten Unterschiede zwischen den Zementen. Die Ergebnisse können in der nur geringen oberflächlichen Lokalisation von Kupfer im kupfermodifizierten Zement begründet liegen.de
dc.description.abstractengAIM The aim of this in vitro study was to evaluate the biocompatibility and the antimicrobial effect of a copper-loaded zinc phosphate cement in comparison to a conventional unloaded reference cement. MATERIALS/METHODS Two materials were compared in this study (Hoffmann´s Copper Cement and Hoffmann´s Cement, Hoffmann Dental Manufaktur, Germany). Solid specimen were used. The tests started with measurements of the arithmetic surface roughness, the free surface energy, and the surface allocation of copper. Microbial testing included the incubation with Streptococcus sanguinis and Candida albicans followed by fluorometric assays. Cytotoxicity-testing was performed with an CCK8 assay on L929 mouse fibroblasts and GF1 human gingiva-fibroblasts. The tests closed with imaging via light and raster electron microscopy. RESULTS There were no relevant significant differences in surface roughness, surface free energies, cytotoxicities or antimicrobial activities. However both materials showed high cytotoxicities. The surface allocation of copper in the copper-loaded cement was low. CONCLUSION The copper-additive did not exhibit any significant antimicrobial potential though a high cytotoxicity was shown. Additionally, all other tested parameters did not show any relevant significant differences within the two cements. These findings may result from the low amount of copper in the copper-loaded phosphate cement.de
