dc.description.abstractengNephrolithiasis is a major health problem caused by hyperoxaluria. The sulfate anion transporter-1 which transports sulfate and oxalate might be involved in the development of hyperoxaluria, so further information concerning the influence of oxalate and its precursors on the expression of sat-1 is needed. Sat-1 expressing caco2 cells taken from a human adenocarcinoma were used to demonstrate the interaction of oxalate and its precursors glyoxylate, glycolate, glycine and hydroxyproline with sat-1. Caco2 cells were incubated with one of these substances for four days. After incubation changes in sat-1 mRNA expression were detected by using real-time polymerase chain reaction while protein expression of sat-1 was examined by Western blotting.Oxalate and its precursors glyoxylate, glycolate und hydroxyproline stimulated the expression of sat-1 mRNA while glycine did not. Oxalate and glyoxylate caused the most significant stimulation so that further examinations concerning the adaquate time of incubation and concentration of oxalate and glyoxylate were conducted.de
