dc.description.abstractengThe production of EHM with undifferentiated murine embryonic stem cells was improved and stabilized by multiple factors (Matrigel-free culture, supplementation of ascorbic acid, culture conditions). Cardiogenesis of undifferentiated mES cells in EHM culture or EB culture shows great similiarity during development. Concerning the maturation of the heart tissue the EHM culture proves to be cleary advanced compared to the EB culture. Compared to native myokardium EHM seem to be similar to perinatal myokardium. This work was able to show that EHM have a similar adjustment response to mechanical load as murine myokardium (transcription, contraction). Forcing mechanical load on undifferentiated ES cells lead to homogenization of the heart tissue by aligning the cells along the force gradient. Overloading on the other hand lead to functional tissue damage. In summery the data shows that mES EHM prove to be a good solution for exploration of mechanisms of cardiogenesis and the influence of biomechanical load to the heart.de
