dc.description.abstractengDDR1 is an ubiquitous cell surface receptor that binds collagens and can be activated by them. The DDR1-knockout-mouse develops osteoarthritis in the TMJ cartilage that then demonstrates an unstructured fibre apparatus. This knowledge gave reason for this thesis which, for the first time, examines dental and periodontal tissues of the DDR1-knockout-mouse histologically and compares them to those of the wildtype-mouse. Between both test groups, there were no differnces in the structural conception of the ECM in the tissues pulp, dentine, cement, PDL, alveolar and jaw bone respectively. Particular attention was paid to the collagenous fibre network of the PDL. It showed regular development and no sign of pathological processes. This leads to the conclusion that the lack of DDR1 on cell surfaces of oral tissues has no impact on the phenotypic structure. The second part of the thesis novelly examined the expression of the class II fibulins-3, -4 and -5 in oral tissues of the wildtype- and the DDR1-knockout-mouse. Fibulins are important structural proteins in the ECM, class II fibulins are primarily associated with the formation of elastic fibres. Fibulin-3, -4 and -5 were detectable in the ECM of pulp, crown dentine, PDL and pericellular areas of cement, alveolar and jaw bone lacunae respectively. In the matrix of cement only fibulin-4 could be found. Expression of the examined fibulins in the DDR1-knockout-mouse was consistent for the most part, nontheless, in dentine and cement matrix variations were detectable. These differences point to a biochemical variation in the wildtype- and the DDR1-knockout-mouse in spite of a structural comparability. This thesis shows that fibulin-3, -4 and -5 are widespread in the ECM of oral tissues and variable in their expression. Moreover, besides a role within elastic fibres, the demonstrated results point to a yet unknown role of the examined proteins in the ECM formation outside the elastic fibre system.de
