dc.description.abstractengThe aim of the present animal study entitled "bone regeneration in vivo by the combined release of BMP and VEGF from PDLLA / CaCO3 composite scaffolds" was an improvement of bone regeneration by Mandibuladefekten by the combined release of osteoinductive proteins BMP and VEGF from PDLLA / CaCO3 composite scaffolds. For this PDLLA / CaCO3 composite scaffolds with concentrations of 24 µg, 48 µg and 96 µg BMP and 0.24 µg, 1.5 µg and 6 µg VEGF were each combined and loaded in Critical size defects of the mandible of adult rats. After the observation periods 4 weeks, 12 weeks and 26 weeks, the histological examinations were performed. The newly formed bone fraction was determined by computer-assisted histomorphometry and the results were analyzed by analysis of variance (ANOVA) statistically. Compared to the single dose of BMP bone regeneration can be enhanced by combining 24 µg BMP and 1.5 µg or 6 µg VEGF particularly in the long-term monitoring (p <0.05). In addition, could be detected comparable bone formation through the combination of 24 µg BMP and 1.5 µg or 6 µg VEGF after 4, 12 and 26 weeks, as indicated by 48 µg BMP in single dosage. The combined release of BMP and VEGF from PDLLA / CaCO3 composite scaffolds new bone formation compared to the single dose could be increased significantly. Moreover, it was shown that the applied concentrations of BMP can be reduced by the addition of VEGF, without causing a reduction in bone formation rate.de
