dc.description.abstractengOsteoporosis-induced bone fractures and their delayed healing are a frequent and worldwide problem. Thus, safe and non-pharmacological methods for enhancing poor healing tendency in osteoporotic bones are of high medical interest. In this experiment the effect of mechanical stimulation (vertical Whole-Body Vibration) on metaphyseal tibial bone healing in intact and ovariectomized rats was investigated. Half of 60 female Sprague-Dawley rats were ovariectomized at the age of three months. Ten weeks later all rats underwent bilateral metaphyseal osteotomy of tibia. Five days after the osteotomy half of the ovariectomized and half of the intact rats were subjected to vertical Whole-Body Vibration (90 Hz, 0.5 mm, 4 × g acceleration) for 15 minutes twice a day during 30 days. The other animals were not exposed to this treatment. After the rats were decapitated one randomly chosen tibia of each rat was used for biomechanical and histological analyses. It showed that vibration did not influence body weight and food intake. In ovariectomized rats cortical and callus densities (97 vs. 99 %, 72 vs. 81 %) and trabecular structure (9 vs. 14 trabecular nodes) were improved while biomechanical properties of tibia were reduced after mechanical stimulation. Callus bridging in ovariectomized rats occurred earlier than in intact rats where bridging was found later after vibration. In conclusion, the applied vibration regimen (90 Hz, 0.5 mm, 4 × g acceleration, 15 min twice a day) showed positive effects on bone tissues but was not optimal for fracture healing.de
