This present work deals with the peroxisomal biogenesis and should help to understand it. The work focuses on the participation of the dynamin-like protein Vps1p in the process of the peroxisomal biogenesis in S. cerevisiae. It is already published that Vps1p is important for division of preperoxisomal structures into individual peroxisomes. The results of the experiment showed that Vps1p has another function in peroxisomal biogenesis; this function is ER dependent and happens already at an early state of peroxisomal neogenesis. Since a connection between peroxisomes and ER has been observed before, another part of this work deals with the local and functional connection of peroxisomes and ER. To investigate this, there are shown live-cell experiments and also a protocol for immunofluorescence microscopy has been established. Both experiments showed clearly the connection between the two organelles. Because of that a lifetime commitment between peroxisomes and ER is most likely. Another aspect dealt with the question whether peroxisomes in wild-type cells - as currently postulated in the literature - arise only by the way of growth and division or via de novo formation. The results of this study indicate that in wild-type cells de novo formation of peroxisomes takes place. It is possible that both systems run in parallel in wild-type cells.
