dc.description.abstractengAtherosclerosis is among the leading causes of death in most parts of the world. The aetiology of this wide-spread, multifactorial disease is not yet entirely understood. Identified conventional risk factors include smoking, diabetes mellitus, hypercholesterolaemia and hypertension, but genetic influence is also proving to be an important risk factor. It has been reported that single nucleotide polymorphisms (SNP) in brap are associated with an aggravation of the atherosclerotic burden in certain human populations. Further evidence indicates that one of them leads to an increased expression of BRAP. The aim of this thesis, therefore, was to examine the effects of a reduced expression of BRAP on the development of atherosclerotic plaques in ApoE-knockout mice. For this purpose ApoE-deficient C57BL/6J-mice (ApoE-/-) with a heterozygous knockout of BRAP (IMP+/-) were set on either a high fat diet or a standard diet for 6 months. ApoE-/- IMP+/+ mice set on the same regime served as a control group for the respective diet. All mice were phenotyped and the atherosclerotic lesions at the aortic root as well as in the brachiocephalic artery were measured. Further examination was carried out by immunohistochemial analysis followed by an evaluation for plaque instability. The results indicate that a reduced expression of BRAP in ApoE-/- is not followed by a lesser extent of atherosclerosis. However, ApoE-/- IMP+/--mice show larger abdominal fat pads and have a lower LDL/HDL ratio. This could be an explanation for the observed increased cardiovascular risk associated with an altered expression of BRAP.de
