dc.description.abstractgerDie vorliegende Arbeit beschäftigt sich mit der molekularen Entstehung von Thymusneoplasien. Thymusneoplasien sind sehr seltene Neubildungen im Thymus, die aus dem Epithel des im Mediastium situierten Thymus hervorgehen. Über ihre Ätiopathogenese ist wenig bekannt, sie stellen jedoch die adulte Tumorentität mit der geringsten Mutationslast dar. Eine zielgerichtete Therapie der auch nach Jahren zu Lokalrezidiven neigenden Tumore ist daher schwierig. In neueren Studien konnte gezeigt werden, dass in einem Anteil der Thymustumore eine Deletion des Apoptose-Enzyms Caspase-3 vorliegt, was mit einer verbesserten Prognose einhergeht. Dem klassischen Rollenverständnis der Apoptose in neoplastischen Erkrankungen widerspricht dies zunächst, jedoch ergaben zahlreiche Publikationen in der jüngeren Zeit Hinweise auf non-kanonische Funktionen der Caspase-3, die zu einer Tumorprogression beitragen können. Eine dieser non-kanonischen Funktionen ist die Interaktion mit dem transkriptionellen Co-Aktivator YAP, welcher zahlreiche Funktionen in der Regulation der Gewebehomöostase physiologischer und neoplastischer Zellverbände erfüllt. Es wurde anhand zweier etablierter thymischer Zellmodelle der Einfluss der Modulation des Caspase 3-Status in vitro untersucht. Hierfür wurden zunächst erfolgreich Zellmodelle erzeugt und etabliert. Es folgten Wachstumsanalysen in 2D- und 3D-Zellkultur, die in der Thymuskarzinomzelllinie 1889c einen wachstumssteigernden Effekt, in der Thymom-Zelllinie TAB-1 einen wachstumshemmenden Effekt der Caspase-3 nachweisen konnte, was die Kontextsensitivität der Caspase-3-Effekte anschaulich darstellt. In nachfolgenden Analysen wurde der YAP-Status in den Zellen analysiert, wobei sich, konsistent mit den beobachteten Effekten, keine differenzielle Regulation von YAP im TAB-1-Modell zeigte. Im 1889c-Modell konnte eine erhöhte Lokalisation von YAP im Zellkern, die mit einer Aktivität des Proteins korreliert ist, fluoreszenzmikroskopisch nachgewiesen werden. Eine im Anschluss erfolgte Erzeugung und Etablierung eines stabilen YAP-Knockdown-Modells in 1889c-Zellen konnte die in 1889c beobachtete Verminderung der Klonalität durch Caspase-3 partiell reproduzieren. Um die in vitro generierten Ergebnisse zu validieren, wurden immunhistochemische Untersuchungen in Bezug auf die YAP-Expression an Gewebeproben von 111 Patient*innen mit epithelialer Thymusneoplasie durchgeführt, wobei sich zeigte, dass eher die dem Normalthymus ähnlichen Subtypen eine hohe Expression von YAP aufwiesen als die high-grade Neoplasien, was nicht die sonst häufig beobachtete Korrelation von YAP mit fortgeschrittenem Tumorstadium und schlechter Prognose reflektiert. Eine Abhängigkeit des Gesamtüberlebens vom YAP-Status konnte nicht gezeigt werden. Nebenbefundlich zeigte sich in Normalthymi eine starke YAP-Expression in Typ-I-Epithelzellen. Die Ergebnisse deuten nicht auf eine dominierende Rolle von YAP in Thymusneoplasien hin. Die in vitro generierten Ergebnisse deuten zwar auf eine Rolle der Caspase-3 und von YAP hin, einer Validierung im Gewebe hielten sie jedoch nicht stand. Eine Rolle des CASP3-YAP-Moduls im Kontext der Onkogenese scheint jedoch möglich, sollte aber in anderen Entitäten und auch in vivo validiert werden. Auch ob YAP eine physiologische Rolle in Thymusentwicklung und -involution spielt, könnte eine Fragestellung für zukünftige Untersuchungen sein.de
dc.description.abstractengThymic neoplasms are very rare neoplasms in the thymus that arise from the epithelium of the thymus, located in the mediastinum. Little is known about their etiopathogenesis, but they represent the adult tumor entity with the lowest mutational burden, therefore a targeted therapy is difficult. Recent studies have shown that there is a subset of cases in which a deletion of the apoptosis enzyme caspase-3 is occurring, which seems to be associated with an improved prognosis. This is contradictory to the classical understanding of the role of apoptosis in neoplastic disease, but numerous publications have recently shown non-canonical functions of caspase-3, which can contribute to tumor progression. One of these non-canonical functions is the interaction with the transcriptional co-activator YAP. YAP fulfills numerous functions in the regulation of tissue homeostasis in physiological as well as neoplastic tissues. In this thesis, the influence of the modulation of the caspase-3 status was examined in vitro using two known thymic cell models. For this purpose, cell models were first successfully created and established. This was followed by growth analysis in 2D and 3D cell culture systems. A growth-enhancing effect of caspase-3 in the thymus carcinoma cell line 1889c and a growth-inhibiting effect of caspase-3 in the thymoma cell line TAB-1 could be shown. This points to the context sensitivity of the caspase-3 effects. In subsequent analyses, the YAP status in the cells was analysed. It was found to be consistent with the observed effects on cellular growth and showed no differential regulation of YAP in the TAB-1 model. In the 1889c model, an increased localization of YAP in the cell nucleus could be detected by fluorescence microscopy which is correlated with an activity of the protein. Subsequent generation and establishment of a stable YAP knockdown cell model in 1889c cells was able to partially reproduce the reduction in clonality observed in 1889c by caspase-3. To validate the results generated in vitro, immunohistochemical studies of the YAP expression were performed in tissue samples from 111 patients with epithelial thymic neoplasms. It was possible to confirm that low-grade tumors exhibiting histomorphologic traits of the normal thymus, have a higher YAP expression compared to high-grade neoplasms like thymic carcinoma. This is incompatible with the otherwise frequently observed correlation of YAP expression with advanced tumor stage and poor prognosis. A dependency of the overall survival on the YAP status could not be shown. An additional observation was the strong YAP expression in type I epithelial cells of normal thymi. The results do not support a dominant role of YAP in thymic neoplasia. Although the results generated in vitro indicate a role for caspase-3 and YAP, they have not withstood validation in tissue. However, a role of the CASP3-YAP module in the context of oncogenesis seems possible. However, it should be validated in other entities and in vivo. Also, whether YAP plays a physiological role in thymic development and involution remains to be elucidated.de
