dc.description.abstractgerDie häufigste hereditäre Neuropathie, die Charcot-Marie-Tooth-Erkrankung Typ 1A, ist durch eine Duplikation des für das peripheral myelin protein 22 (PMP22) codierenden Gens bedingt. Trotz jahrzehntelanger Forschung ist die Funktion des Transmembranproteins PMP22 und damit einhergehend der pathophysiologische Mechanismus der Erkrankung nach wie vor nicht endgültig entschlüsselt und es gibt bislang neben allgemeinen Therapiemaßnahmen nur wenig klinisch erprobte Wirkstoffe zur Behandlung. Um die biologische bzw. pathophysiologische Funktion eines Proteins zu entschlüsseln und somit neue Therapieansätze zu begründen, ist u. a. die Untersuchung der Protein-Protein-Interaktionen des Zielproteins sinnvoll. Im Rahmen dieser Arbeit wurde daher ein Protokoll zur Identifizierung von In-vivoInteraktionspartnern von PMP22 im peripheren Nerven der Ratte entwickelt, welches auf Immunopräzipitation und anschließender Massenspektrometrie basierte. Um den Einsatz denaturierender Detergenzien zur Anreicherung des Transmembranproteins PMP22 vor der Immunopräzipitation zu ermöglichen und um transiente sowie schwache ProteinProtein-Interaktionen von PMP22 zu bewahren, wurde ein Proteinquervernetzungsschritt zur kovalenten Verknüpfung von PMP22 und dessen benachbarten Proteinen etabliert. Neben der Proteinquervernetzung wurde die Solubilisierung für PMP22 und die Immunopräzipitation selbst mit und ohne Proteinquervernetzung optimiert. Hierbei zeigten die verwendeten Detergenzien unterschiedliche Wirksamkeit und Natriumdodeylsulfat-Polyacrylamid war als einziges Detergenz in der Lage, PMP22 aus proteinquervernetzten Homogenisaten zu solubilisieren. In nicht-proteinquervernetzten Proben zeigte Dodecyl-β-D-maltosid das beste Solubilisierungsergebnis. Die Immunopräzipitationsprotokolle wurden durch Optimierung des Einsatzes von AntikörperMengen und Dynabeads etabliert. Die durch die optimierten Immunopräzipitationsverfahren gewonnenen Proben aus Ratten-Ischias-Nerven wurden dann massenspektrometrisch vermessen, um erstmalig das gesamte Interaktom von PMP22 zu untersuchen. Die Analyse der probatorischen Massenspektrometrie-Ergebnisse legte eine Funktion von PMP22 bzw. dessen Interaktionspartnern für Zell-Zell-Adhäsionen, FokalAdhäsionen und das Aktin-Zytoskelett sowie für die Regulation intrazellulärer Signalkaskaden nahe. Konkret wurden u. a. myelin protein zero, Periaxin, 14-3-3- Proteine und das Aktin-bindende Protein Cofilin-1 spezifisch in den PMP22- Immunopräzipitationen angereichert. Interaktionen von PMP22 mit myelin protein zero Zusammenfassung 96 und Perixaxin könnten die Funktion von PMP22 als Myelinarchitektur-Stabilisator erklären, während die Interaktion von PMP22 mit 14-3-3-Proteinen und/oder Cofilin-1 die molekularen Korrelate der bei der Charcot-Marie-Tooth-1A-Erkrankung gestörten Schwann-Zell-Ausrichtung, Formgebung und Polarisierung in der Entwicklung des Ischias-Nerven darstellen könnten. Mithilfe des im Rahmen dieser Arbeit etablierten Protokolls und der für PMP22 etablierten Proteinquervernetzung sollte es in folgenden Arbeiten möglich sein, die hier diskutierten Interaktionspartner zu validieren und somit neue Therapieansätze zur Behandlung der Charcot-Marie-Tooth-1A-Erkrankung zu begründen.de
dc.description.abstractengThe most common hereditary neuropathy, Charcot-Marie-Tooth disease type 1A (CMT1A), is caused by a duplication of the gene coding for the peripheral myelin protein 22 (PMP22). Despite long lasting scientific investigations, the function of this transmembrane protein and thereby pathophysiological mechanisms of the disease remain obscure and treatment options are limited. In order to elucidate the physiological and pathophysiological function of PMP22 and thereby suggest new treatment approaches, the investigation of its protein-protein interaction partners is meaningful. In the given work, we established an immunoprecipitation mass-spectrometry approach to identify in-vivo protein-protein interaction partners of Pmp22 in the sciatic nerves of rats. In order to allow the application of denaturing detergents and conserve also transient and low-affinity interactions we employed protein crosslinking covalently linking Pmp22 to the proteins in close proximity. We optimized the solubilization of Pmp22 in the sciatic nerve and the immunoprecipitation of Pmp22 from the nerve homogenate with and without crosslinker. The detergents applied in this work were differently effective in solubilizing Pmp22. In crosslinked homogenate, Pmp22 could be solubilized only using sodium dodecyl sulfate-polyacrylamide while non-crosslinked samples were most effectively solubilized with dodecyl-beta-D-maltoside. The immunoprecipitation protocol was further successfully optimized in terms antibody and dynabead amounts. As a pilot experiment, we analyzed the immunoprecipitation eluate using liquid chromatography tandem mass-spectrometry investigating the potential interactom of Pmp22 in rats’ sciatic nerves. The analysis of this pilot experiment considering (1) specific enrichment and (2) Gene Ontology terms suggested a potential function of Pmp22 and its protein interaction partners in cell-cell adhesions, focal adhesions, the actin cytoskeleton and intracellular signaling pathways. Namely, myelin protein zero, periaxin, 14-3-3 protein and the actin-binding protein cofilin-1 were among others specifically enriched in the Pmp22 immunoprecipitation eluate. Interactions of Pmp22 with myelin protein zero and periaxin could explain the function of Pmp22 as a stabilizer of the myelin architecture. The interaction with 14-3-3 and/or cofilin-1 could furthermore represent the molecular correlate of the disrupted shaping, structure and polarity of Schwann cells in the development of peripheral nerves in CMT1A. Within follow-up investigations using the given works’ Pmp22 immunoprecipitation protocol and crosslinking approach, these potential protein interaction partners could be validated thereby paving the way for novel treatment strategies in CMT1A.de
