Abstract
English
Mutations in the beta-synuclein protein have been directly linked to Dementia with Lewy bodies (DLB). In particular, P123H mutation was linked to a familial case of DLB, whereas V70M mutation was identified in a sporadic case of the disease. Previous work done in overexpression studies have shown that these two mutants might exert their neurotoxicity through different cellular pathways: V70M affecting mitochondrial fitness and P123H impairing spontaneous neuronal activity. The objective of this thesis was to gain more insight into how these proteins exert such different effects. Results show that while these proteins interact with the same affinity with biologically relevant membranes, their neurotoxicity might relay on their different aggregation propensities.
