dc.description.abstracteng | Falcarin-type polyacetylenes (PA) are a group of putative plant defense metabolites produced by carrot plants. However, the elucidation of PA biosynthetic pathway and the biological function of PA in carrot plants are not yet fully understood. This research first investigated the influence of endogenous PA on plant resistance against two fungi pathogens, Mycocentrospora acerina and Botrytis cinerea, limited inhibition effects of PA were observed. Secondly, strong quantitative trait loci (QTLs) for PA accumulation in carrots were identified, which co-located with several candidate fatty acid desaturase 2 (FAD2) and eceriferum 1 (CER1) genes. Meanwhile, spatial and temporal gene expression studies of candidate FAD2 genes by qRT-PCR showed the high activities of these genes in lateral roots and young plants. Lastly, the function of a FAD2 and a CER1 candidates was characterized respectively through loss-of-function analysis and revealed the altered PA levels and FAD2 gene co-expression pattern. This research has provided insights into PA as putative plant defense metabolites and the candidates of FAD2 genes that involve in PA biosynthetic pathway. | de |
