Abstract

English

As part of this study, changes in the neuronal redox balance in the brains of Rett mice are mapped and correlated with the progression of Rett syndrome. Fluorescence microscopy techniques (primarily ratiometric CCD camera imaging and two-photon microscopy) were used along with the optical redox indicator roGFP1. This redox indicator is already stably expressed in the Rett mice, allowing for optical analyses to be performend directly on acutely isolated brain tissue without the need for prior transfection or transduction. With this method it was possible to document the changes in the neuronal redox balance in different brain areas, in different ages and genotypes of mice.
