dc.description.abstractgerOsteoporose und Sarkopenie sind ein weltweites Gesundheitsproblem, welches insbesondere die alternde Bevölkerung in den Industrieländern betrifft. Strontiumranelat ist ein Osteoporosemedikament, dessen Effekt auf das Muskelgewebe bisher ungeklärt ist. Da die Integrität des Muskelgewebes, insbesondere da häufig Sarkopenie und Osteoporose gemeinsam auftreten, immensen Einfluss auf die Integrität des Knochens hat, ist es relevant, ob Strontium auf die Muskulatur wirkt. In unserem Versuch wurden dazu 60 drei Monate alte Sprague-Dawley ovarektomiert (ovx, n=48) oder nicht ovarektomiert (non-ovx, n=12). Nach acht Wochen entwickelten die ovarektomierten Ratten eine Osteoporose und alle Tiere wurden bilateral an den Metaphysen der Tibiae osteotomiert. Die Tiere wurden in fünf Gruppen eingeteilt: non-ovx, ovx und drei Gruppen die Strontiumranelat in einer Dosierung von 10,5g/kg Futter erhielten. Die Strontiumgruppen erhielten entweder vor der Osteotomie (SR vor Osteo) nach der Osteotomie (SR nach Osteo) oder vor und nach der Osteotomie (SR vor und nach Osteo) Strontiumranelat mit dem Futter sodass sich eine durchschnittliche Aufnahme von 654mg/kg Körpergewicht pro Tag ergab. Fünf Wochen nach Osteotomie wurden die Tiere euthanasiert und obduziert. Es wurden der M. Gastrocnemius, der M. Soleus und der M. Longissimus entnommen, unterschiedlich gefärbt und die Anzahl an Zellen, Kapillaren und das Verhältnis von Kapillaren zu Zellen bestimmt. Des Weiteren wurden der Muskelfaserquerschnitt und das Verhältnis von glykogenolytischen zu oxidativen Muskelfaserzellen untersucht. Die meisten Effekte, die Strontiumranelat auf die Muskulatur der osteopenen Ratte in diesem Versuch zeigte, lassen sich als Folge des Östrogenmangels aufgrund der Ovarektomie interpretie- ren, allerdings mit der Ausnahme des Verteilungsmusters von glykogenolytischen und oxidativen Fasern beim M. Longissimus. Beim M. longissimus war der Quotient aus oxidativen und glykogenolytischen Zellen bei der der SR vor und nach Osteo gegenüber der ovx- Gruppe si- gnifikant vermindert, die Relevanz des Überwiegens der glykolitischen Fasern in dieser SR Gruppe bleibt unklar. Es konnte kein negativer Einfluss der Strontiumranelatgabe auf die Muskulatur festgestellt werden, sodass es vor dem Hintergrund, dass das Strontiumranelat die Knochendichte bei dem längsten Verabreichungszeitraum am stärksten erhöhte, ohne negative Effekte auf die Frakturheilung zu haben, sich die Langzeitgabe empfiehlt.de
dc.description.abstractengOsteoporosis and sarcopenia are a global health problem affecting in particular the aging population in industrialized countries. Strontium ranelate is an agent for osteoporosis therapy. It is still unclear what effect Strontium has on muscle tissue. Since the integrity of muscle tissue has a tremendous impact on the integrity of the bone, especially as sarcopenia and osteoporosis commonly occur together, it is relevant whether strontium affects the muscles. In our clinical trial 60 three-month-old Sprague-Dawley either ovariectomized (ovx, n = 48) or not ovariectomized (non-ovx, n = 12). After eight weeks, the ovariectomized rats had developed osteoporosis and all animals were osteotomized bilaterally to the metaphyses of the tibia. The animals were divided into five groups: non-ovx, ovx and three groups receiving strontium ranelate at a dose of 10.5 g/kg of food. The strontium groups received strontium ranelate with the diet either before osteotomy (SR before osteo) after osteotomy (SR after osteo) or before and after osteotomy (SR before and after osteo). The average intake of Strontium ranelate accumulated to 654 mg/kg body weight per day respectivly. Five weeks after osteotomy, the animals were euthanized and autopsied. The M. gastrocnemius, the M. soleus and the M. longissimus were stained individually and the number of cells, capillaries and the ratio of capillaries to cells were determined. Furthermore, the muscle fiber cross section and the ratio of glycogenolytic to oxidative muscle fiber cells were studied. Most of the effects of strontium ranelate on the muscles of the osteopenic rat in this experiment can be interpreted as a consequence of estrogen deficiency due to ovariectomy, with exception being the distribution pattern of glycogenolytic and oxidative fibers in the M. longissimus. In M. longissimus the ratio of oxidative to glycolytic cells in the SR before and after osteo group was lower compared with that in the ovx group. The relevance of the predominance of glycolytic fibers in this SR group remains unclear. We found the strontium ranelate dose had no negative influence on the muscle tissue. The highest bone density was achieved over the longest administration period of strontium ranelate, without having adverse effects on fracture healing. Therefore a long-term administration is recommended.de
